Objective:To study the relationship between the SNP ofβ2-AR gene 5'-regulatory region and essential hypertension (EH) in Xinjiang Kazak and Uygur patients; by comparison with the Uygur people to explore the genetic susceptibility of EH Mechanism in different ethics.Methods:(1)Polymerase Chain Reaction (PCR) and single-base extension and MALDI-TOF-MS(Matrix assisted laserdesorption ionization time of flight mass spectrometry) combination techniques were used to detectβ2-AR gene 5'-regulatory region of the 6 loci SNP in 150 Xinjiang Kazak EH patients and 150 controls, and 100 Xinjiang Uygur EH patients and 100 controls.Biochemical analyzer was used to detect lipid and other related biochemical parameters.(2)SPSS16.0 statistical software was used for statistical analysis to calculate genotype frequencies of the two groups to confirm the consistent with Hardy-Weinberg equilibrium;χ2 test was used to the compare with groups allele frequencies and allele frequencies; t test was used to compare the two clinical indicators;Logistic regression was used for EH risk factors; SHEsis and other.software were used to analyse linkage disequilibrium and haplotype.Results:(1)There were gene polymorphisms ofβ2-AR gene 5'-regulatory region of six loci rs2053044,rs17108803,rs12654778,rs11168070,rs11959427,rs2895795 in the Xinjiang Kazak and Uygur populations.No difference was detected of rs2053044 locus genotypes GG,GA, AA and allele G, A frequency in the EH group and control group between ethnic groups(P>0.05);No statistically significant was detected of rs12654778,rs2895795, rs17108803,rs11959427 locus of all genotype and allele frequencies of EH in the Kazak and Uygur ethnic group and the normal control group in the distribution of two inter-ethnic difference (P> 0.05).Two significant inter-ethnic differences were detected of rsl 1168070 locus genotypes CC,CG,GG and allele C,G frequency in the EH group and control group (P<0.05);(2)There is a strong linkage disequilibrium in the Kazak EH group and control group in both population the Kazak population do linkage disequilibrium and haplotypes analysis. There is a strong linkage disequilibrium coefficient of rsl 1168070,rs2053044, rs2895795 polymorphism loci D'0.9122,1.000,0.7689,respectively,the correlation coefficient r2,respectively 0.1273,0.1606,0.3900, normal control group,rs11168070, rs12654778, rs17108803 gene polymorphism is linkage disequilibrium. Haplotypes GTCCAT,GACTGT,ATGCGT,ATCTGT,ATGTGT,GTCCGT,GTCTAT,GACCAT,GTCT GT associated with EH, which GTCCAT,GACTGT,ATGCGT are risk factors, while the ATCTGT,ATGTGT,GTCCGT,GTCTAT,GACCAT,GTCTGT is a protective factor.(3)The systolic blood pressure, diastolic blood pressure, mean arterial pressure, body mass index, total cholesterol of Kazak EH group were significantly higher than Uygur EH group, overweight, obesity, abdominal obesity, elevated blood glucose,lipid metabolic disorders are two common national EH risk factors. In addition, too little intake of vegetables is also a risk factor for Kazak EH people, but has nothing to do with the Uyghrs.Conclusions:(1)Kazak and Uighur groups in Xinjiangβ2-AR gene 5'-regulatory region of five loci rs2053044,rs17108803,rs12654778,rs11959427,rs2895795 genetic polymorphism exists,its genotype and allele frequency distribution in the EH group and normal control group,no difference between the two peoples;(2)rsl 1168070 locus genotype and allele frequency distribution in the two national EH group and normal control group, there are significant differences.(3)Kazak population rs2053044,rs17108803,rsl2654778,rs11168070, rs11959427,rs2895795 polymorphic loci there is a strong linkage disequilibrium, which were composed of six haplotypes GTCCAT,GACTGT,ATGCGT Xinjiang Kazak EH risk factors, while the ATCTGT,ATGTGT,GTCCGT,GTCTAT,GACCAT,GTCTGT is a protective factor.
