| Objective:To detect the expression of PIRH2 in chronic cervicitis, cervical intraepithelial neoplasia(CIN) and cervical carcinoma, exploring the correlation between the expression of PIRH2 and the clinical pathological characteristics of cervical carcinoma.Methods:Seventy-three cases of cervical carcinoma,43 cases of cervical intraepithelial neoplasia(CIN) and 13 cases of chronic cervicitis were selected randomly from those who received primaly diagnosis and treatment in the Department of Gynecology and Obstetrics of the second Xiang ya Hospital of Central South University, Hunan Province Maternal and Child Health Hospital, HuNan Province Tumor Hospital from January 2007 to August 2009. There were 16 cases of CIN I,13 cases of CINâ…¡,14 cases of CINIII; 64 cases of squamous carcinoma,9 cases of adenocarcinoma; 59 cases of stage I,14 cases of stage II; 14 cases with pelvic lymph node metastasis and 59 cases without pelvic lymph node metastasis. According to histological grade of squamous carcinoma, there were 33 cases of well differentiation (G1),24 cases of moderate differentiation (G2),16 cases of poor differentiation (G3). All tissues were pathologically confirmed and no patients had ever received radiotherapy, chemotheraPy or any other biological therapy before. S-P immunohistochemistry stain of PIRH2 was carried out on all of the samples. The results were analysised in the relationship between the expression of PIRH2 and clinical pathological characteristics. The data were analyzed with statistical software SPSS 16.0. Comparison among multiple groups using Kruskal-Wallis H test, and Comparison between two independent samples of group using Mann-Whitney U test, P<0.05 was considered the level of significance.Results:(1) The positive expression rate of PIRH2 in group of chronic cervicitis, CIN I,CINâ…¡-â…¢, cervical carcinoma was 0,18.75%, 59.26%,68.49%respectively. There was significant difference among four groups statistically (P=0.000 P<0.05).(2) The Positive expression rate of PIRH2 in cervical carcinoma group of stage I a, stage I b, stage II a was 63.33%,68.97%,76.92% respectively. There was no significant difference among three groups statistically (P=0.601, P>0.05).(3) The Positive expression rate of PIRH2 in cervical carcinoma of differentiation G1 group, G2 group, G3 group was 54.55%,75.00%, 87.57%respectively. There was significant difference among three groups statistically (p=0.049 P<0.05).(4) The Positive expression rate of PIRH2 in cervical carcinoma with pelvic lymph node positive group and pelvic lymph node negtive group was 92.85%,62.71%respectively, There was significant difference between two groups statistically (P=0.020 P<0.05).(5) The Positive expression rate of PIRH2 in cervical carcinoma group of squamous carcinoma and adenocarcinoma was 67.18%,77.78% respectively. There was no significant difference between two groups statistically (P=0.525 P>0.05). Conclusion:(1) PIRH2 expresses in chronic cervicitis, cervical intraepithelial neoplasia (CIN) and cervical carcinoma in cellular nucleolus with an ascending tendency.(2) PIRH2 expression also has relevance with tissue differentiation and Pelvic lymph node metastasis, there was no significant correlation between PIRH2 expression and cervical carcinoma of clinical stage ages and the diameter of lump. Above all, our experiment indicated that the protein PIRH2 may play an important role in cervical squamous carcinoma infiltration and metastasis. |
