【Objective】 This experiment mainly to study the expression of Pirh2 and Her-2 protein in differentiated thyroid carcinoma and benign thyroid disease tissues,and analysis the relationship between Pirh2 and Her-2 protein with clinical pathological characteristics of the patients,and explore the correlation with the occurrence,development and prognosis of differentiated thyroid carcinoma.【Methods】 1.100 cases of paraffin embedding specimens collected from January 2012 to December 2013 in the First Affiliated Hospital of Hunan Normal University,including 50 cases of differentiated thyroid carcinoma, 50 cases of benign thyroid disease tissues,this experiment used immunohistochemistry S-P dyeing method to detect the Pirh2 and Her-2 protein expression in differentiated thyroid carcinoma and benign thyroid disease tissues. 2.We collected 60 cases of differentiated thyroid carcinoma patients fresh thyroid tissue specimens in the First Affiliated Hospital of HunanNormal University dairy a surgical operation excision from 2014 June to 2015 February.Postoperative pathology proved 26 cases of differentiated thyroid carcinoma, 34 cases of benign thyroid disease.And we used fresh frozen tissue for Western-blot experiment,It was used to detect the expression of the protein level in Pirh2 and Her-2 protein in two kinds of tissues,and analyze Pirh2 and Her-2 expression in differentiated thyroid carcinoma and benign thyroid disease relationship.The experimental results used SPSS 19.0 software for statistical analysis.【Result】 1.Pirh2 and Her-2 expression in differentiated thyroid cancer and benign thyroid disease that used immunohistochemical method The result which used Immunohistochemistry method showed Pirh2 was mainly localized in tumor cell plasma,a small amount of coloring in the nucleus,brownish yellow.Her-2 was mainly localized at the tumor cell membrane,a few in the cytoplasm,brown yellow.The positive rate of Pirh2 and Her-2 in 50 cases of differentiated thyroid carcinoma were 72%(36/50),44%(22/50),which higher than benign thyroid disease 44%(22/50),4%(2/50)expression significantly(P<0.05). 2.Relationship between expression of Pirh2 and Her-2 in differentiated thyroid carcinoma and the clinical pathological parameters In 50 cases of differentiated thyroid carcinoma,the expression of Pirh2 with age,gender,regional lymph node metastasis,TNM pathologicalstage showed no significant difference(P>0.05).And the expression of Her-2 with lymph node metastasis had significant difference(P<0.05),and age,sex,TNM pathological stage showed no significant difference(P>0.05). 3.The relationship between the expression of Pirh2 and Her-2 in differentiated thyroid carcinoma The relationship between the expression of Pirh2 and Her-2 in differentiated thyroid carcinoma tissues was closed.There were 19 cases of Her-2 positive expression which in 36 cases of Pirh2 positive expression of differentiated thyroid carcinoma;There were 19 cases of Pirh2 positive expression which in 22 cases of Her-2 positive expression of differentiated thyroid carcinoma.By using Spearman correlation analysis,rank correlation coefficientr=0.363, p=0.01,the difference was statistically significant,that Pirh2 is positively correlated with the expression of Her-2 protein. 4.Western-blot was used to detect the expression of Pirh2 and Her-2 in differentiated thyroid carcinoma and benign thyroid disease. The relative expression strength of Pirh2 protein in differentiated thyroid carcinoma is0.535±0.134,higher than in benign thyroid disease in the relative expression intensity of 0.395±0.079,significantly(P<0.05).The relative expression strength of Her-2 protein in differentiated thyroid carcinoma is 0.493±0.127,highethan in benign thyroid disease in the relative expression intensity of 0.405±0.075,significantly(P<0.05).【Conclusion】 1. The high expression of Pirh2 and Her-2 showed in differentiated thyroid carcinoma,and positive correlation,but thyroid benign disease showed low expression.Suggesting that Pirh2 and Her-2 may be an oncogene which play an important role in the occurrence and development of differentiated thyroid carcinoma. 2. The high expression of Her-2 protein in regional lymph node metastasis in patients with differentiated thyroid carcinoma,suggesting that Her-2 may have relevant with growth,invasion or metastasis of differentiated thyroid carcinoma.
