Objective:Research on the phase characteristic and the mechanism of acupuncture on the circadian rhythm disturbance produced by 10 days of serial 8-hour advances of light/dark cycle every 2 days.Method:Syria hamsters were synchronized to standard lighting conditions of LD14:10,and then circadian rhythm disturbance models were produced by 10 days of serial 8-hour advances of light/dark cycle every 2 days. Comparing the influences of acupuncturing those "Baihui(DU20)", "Changqiang(DU1)"points after advanced light/dark cycle on different time. Meanwhile, the expressions of Perl mRNA in SCN and the hypothalamus was observed through the real time RT-PCR at ZT4.Results:After advanced light/dark cycle, the spontaneous behavior of golden hamsters shifted forward. Their average shift extent of different batches and different groups was from 156.14 to 586.00 min. When acupunctured at ZT16 and ZTO, the time of resynchronization was less than the natural recovery group, but they weren't different in statistics. Compared with other groups, the outstanding effective of promoting circadian rhythm resynchronization was observed in acupuncture group at .ZT4, ZT8 and ZT12. While the time of resynchronization of the treated group was more than the natural recovery group at ZT20, but they weren't different with natural recovery group in statistics. At ZT4 acupuncture significantly promoted the expression of Perl mRNA in the SCN and hypothalamus.Conclusions:Firstly, acupuncture could promote circadian rhythm. Secondly, the effective of acupuncture on resynchronization was circadian-dependent effects. In the daytime, the effect was significant, especially in the morning. At man.prim and early night, there was no significant effect. Acupuncture maybe delay the phase recovery at lately night. Last, the mechanism of acupuncture promoting circadian rhythm resynchronization may be that acupuncture could up-regulate the expressions of PerlmRNA in SCN and hypothalamus. |
