| Sugar beet (Beta vulgaris L.) M14 line is the monosomic additional line derived from hybrid between (4×, Beta corolliflora Zoss.) and (2×, B.vulgaris L.) by Dedong Guo et al, which contains the whole No.9 chromosome of Beta corolliflora Zoss.with its successional backcross selection. M14 line showed the apomix character by some evidence.The leaf blade of M14 line was taken as explant on tissue culture in this experiment, then the adventitious bud and adventitious root were successfully induced from the callus respectively, and the plantlets were differentiated. A series of culturing medium were used and selected by orthogonal experimental design. This experiment will establish the rapid proliferous and similar seed-parent heredity of the sugar beet M14 clone, provide platform and establish foundation for apomix genes manipulation. The main contents and results are as follows:1.The optimal callus induced medium was MS+0.1mg/L 6-BA+0.5mg/L 2,4-D+1.5mg/L NAA+1.5mg/L IAA, the callus induced percentage was 83.3%;2.The optimal callus subcultured medium was MS+2.0mg/L KT+1.0mg/L 6-BA+0.3mg/L NAA+1.0g/L Ac, the browning percentage was 5.0%;3.The optimal adventitious bud differentiation induced medium was MS+1.5mg/L 6-BA+1.0mg/L KT+0.3mg/L IAA, the budding inductive rate was 7.5%;4.The optimal adventitious root induced medium was MS+5.0mg/L IBA+1.0mg/L NAA+1.0g/L Ac, the rooting rate reached 72.7%。... |
