Abstrast:As a kind of essential trace elements, Copper involved in many metabolic processes the organism. CP is the auxiliary base composition of many important enzymes and it involved in growth and development and a series of biological process.Ceruloplasmin(Ceruloplasmin, EC1.16.3., abbreviated as CP)is one of the important copper-enzymes in the organism, and people confirmed that Ceruloplasmin is mainly distributed in the animal’s blood,and CP combines more than95%of copper in plasma. Each CP molecule contains6copper atomics. CP can provide available copper to various tissues and cells fast and satisfy the needs of each organ organization for copper. Ceruloplasmin mainly involves in iron’ transport, cell metabolism, free radical scavenging, the development of embryo and the nervous system,the newborn of formation process of blood capillary.Copper sulfate is usually used to prevent the parasitic disease, kill Green clay moss, algae, fungi, etc.the abuse use of copper sulfate lead to the copper accumulation and cause the serious immune system damage, immunity redution and death. As a emergency response protein,CP is closely related to the concentration of Cu2+.And its expression was measured, it can provide the best time and concentration for The rational use of drugs in clinical, and provide one kind of molecular marker for the copper toxicity to Cyprinus Carpio and put forward the theoretical basis.Cyprinus Carpio is one of the important economic fish in north China, so Cyprinus Carpio was used as the research object to study impact of different concentrations of copper to the mRNA expression of Ceruloplasmin, and it has great significance in revealing the toxic effects of copper, the body’s immunity and growth and development of Cyprinus Carpio, at the same time it provides the theory basis for the copper sulfate’s reasonable application in fishery production.This experiment cloned part gene sequence of Ceruloplasmin by RT-PCR method from the organizations of Cyprinus Carpio,about244bp, filled the nucleic acid sequence gap of the protein. Through the contrastive analysis of part gene sequence of Ceruloplasmin i assived and the Ceruloplasmin nucleic acid sequences logined on NCBI,i found the cloned part gene sequence of Ceruloplasmin makes the highest homology with Carassius auratus, reached96%;Aristichthys nobilis (Aristichthys nobilis) reached93%,and the zebrafish (Danio rerio) reached91%. Compared with other species, i found that the homology of Ceruloplasmin nucleic acid sequence is low.It shows that Ceruloplasmin gene must have great differences between species, but the homology is higher in the same species.Under exposured to different concentrations of copper,different organizations of Cyprinus Carpio shows a similar expression pattern by the Semi-quantitative research.The mRNA expression of CP shows an time dependence and dose dependence Under exposured to different concentrations of copper. It makes an increase-to-decline trend followed time,besides the contents of CP mRNA could raise to a highest level but not the same time.Under exposured to different concentrations of copper,there are different degrees of expression between hepatopancreas,brain, kidney, gill and muscle.The research results preliminary reveals that CP should involve in absorption and transport of copper,immunity improved,metabolism regulated and other functions through Cyprinus Carpio exposured to different concentrations of copper.
