China has the highest camellia seed’s production in the world,but a lot of proteins andoil in the camellia seed meal after oil are not into development and utilization, the situationcauses the waste of resources.This paper use reverse micelles system to extract the proteinand oil from camellia seed meal,it has not only high protein extraction rate、simple process,but also avoids the problem of protein denaturation in the traditional extraction.According to the nature of camellia seed meal’s protein, This paper selectes thecationic surface active agent DTAC which is studied less to construct reverse micellarsystem, it has the characteristics of rapid preparation and high protein extraction rate.Research the single factors and interaction effect on the pre-extraction including theDTAC concentration, extraction time, w0value, KCl concentration, sample amount, pHofthe solubilizing water, temperature and so on, the best process of DTAC system pre-extractthe camellia seed meal’s protein by designing the orthogonal experiments are: DTACconcentration is0.06g/mL, w0value of35, the sample amount is0.010g/mL, extractiontime is30min, KCl concentration is0.20mol/L, water pH is7.0, temperature is35℃, theextraction rate of camellia seed meal’s protein reaches88.28%.Based on the single factor experiment of the back-extraction, the best process toback-extract the camellia seed meal’s protein through the Response Suface Methodologyare: pH is6.62, KCl concentration is1.70mol/L, oscillation time is98.50min, the theoryback-extraction rate of camellia seed meal’s protein was87.64%, the real extraction ratewas86.23%. In this experiment, the response surface method model is significant and thefitting is good. The oil phase of back-extraction separate each component by differentphysical properties, the technology is simple and can be recycled.Camellia seed meal’s protein of reverse micelles extraction is through dialysis topurify, then freeze dried to obtain protein products. By studying the protein function, wefound the functional properties in different conditions are different, the development andapplication of functional properties used in the products of amellia seed meal’s protein hasgood prospects for development.
