Objective:We used electroacupuncture which along the visual pathway of on rabbit model of anterior ischemic optic neuropathy (AION),to observe the changes of the optic nerve tissue and apoptosis related factor include Bax and Bcl-2,so as to explore the optic nerve protective effect about electroacupuncture along the visual pathway in ischemia condition and further reveal the possible ways of inhibit cell apoptosis.Method:48 healthy New Zealand rabbits were randomly divided into 4 groups:acupuncture group,electroacupuncture group, model group and blank group, with 12 rabbits in each group. Except the blank group, the other three groups experimental eye (right eye) were droped in compound tropicamide to fully dilated and injected in Rose Bengal 2.5mmol/(ml-kg) via ear-border vein,2/3 of the area above the optic disc was irradiated with a green light with wavelength of 532 nm,power of 75mW,and spot diameter of 500μm for 60s.1 day after modeling,we used the fundus and FFA examination to observe the rabbit optic disc and its surrounding, So as to determine whether the model is successful.then the group of simple acupuncture acupuncture at cuanzhu,yuyao and occipital region points,the group of electroacupuncture increase the electro acupuncture(dilatational wave, frequency of 1Hz strength 5mA) between cuanzhu and occipital region points on the basis of the former. The two groups were were treated 1 times a day,30 minutes each time, continuous treatment for 3 days.Model and blank groups with no intervention.Then each New Zealand rabbits were anesthetized, removed, embedded in paraffin, and observed by HE and immunohistochemical staining.Results:1.HE stain:intervention of 3 days, the structure of the retina of the blank group was complete, and the cells in the optic nerve tissue were normal and orderly, and the retinal ganglion cells were not detached, arranged closely. In model group, the retinal ganglion cell layer hyperplasia,arranged in disorder,decrease in the number,and kernel layer significantly thinner, the number of cells decreased significantly.In acupuncture group, the structure of the retina of the rabbit is basically complete, the retinal ganglion cells are loose, the number is reduced, the number of retinal ganglion cells is missing, the inner core layer becomes thin, and the number of cells is reduced. In electro-acupuncture group,the structure of each layer were basic integrity,the optic nerve tissue cells arranged in order, the inner nuclear layer light thinner and the number of cells decreased,there was a mount of loss in retinal ganglion cells.2 The thickness of the inner layer of retina:the rank means of model group,acupuncture group,electroacupuncture group and blank group were 9.56,18.33,20.83 and 25.28.there was a very significant difference between model group and blank group (P< 0.01). there was no significant difference between the acupuncture group and the model group, there was no significant difference between the electroacupuncture group and the model group.3. Immunohistochemical staining and RGC counts:the Bcl-2 counts of model group,acupuncture group,electroacupuncture group and blank group were 16.33+ 4.583,30.56+7.796,22.78±4.816 and 4.144±10.561, there was a very significant difference between model group and acupuncture group (P=0.000). there was no significant difference between the electroacupuncture group and the model group (P=0.072>0.05). the Bax counts of model group,acupuncture group,electroacupuncture group and blank group were 26.78±11.925,24.78±563,22.89±4.076 and 14.56±5.077, there was no significant difference between the acupuncture group and the model group (P=0.568>0.05). there was no significant difference between the electroacupuncture group and the model group (P=0.270> 0.05). the result of Bax/Bcl-2 of model group,acupuncture group,electroacupuncture group and blank group were 1.64+0.557,0.84±0.254,1.05±0.333 and 0.35±0.091, there was a very significant difference between model group and acupuncture group (P=0.000). there was a very significant difference between model group and electroacupuncture group (P =0.212> 0.05).,there was no significant difference between the acupuncture group and the electroacupuncture group (P=0.212> 0.05).Conclusions:1.Electroacupuncture and acupuncture stimulation along the visual pathway have obvious protective effect on the optic nerve in ischemia condition.2.Electroacupuncture and acupuncture stimulation along the visual pathway can reduce the value of Bax/Bcl-2 in rabbit optic nerve in ischemia condition, but there is no significant difference between the two treatments.3.Electroacupuncture and acupuncture stimulation along the visual pathway may play a role of inhibiting the apoptosis of cells by reducing the value of Bax/Bcl-2 in rabbit optic nerve in ischemia condition.
