Objective: To investigate the effects of diacerein on the experimental rheumatoid arthritis in rats, research the therapeutic effect and mechanism.Methods: We apply fifty male Sprague-Dawley rats as pathological model. The rats were raised for one week, then we could begin the experiment. Totally 8 of 50 rats were selected as control group. 0.1mL of physiological saline was injected into the base of their right hind footpads of each rat by an intradermal injection; 42 of 50 rats were injected in the saline manner by an intradermal injection of 0.1mL of complete Freund's adjuvant to establish the model of adjuvant-induced arthritis. The volume of left hind paws was measured. The levels of serum interleukin1-βwere measured by enzyme linked immunosorbent assay. The levels of serum nitric oxide were measured by colorimetric method. The thymus index was calculated. Made mineral wax sections with left ankle removed from the rats for HE staining to observe the cartilage and synovium.Results: We can see that the intragastric administration of two doses of diacerein can reduce the secondary paws swelling remarkably (P<0.01 or P<0.05) and lessen the pathological changes of secondary arthritis. It can reduce the levels of serum interleukin1-βand serum nitric oxide (P<0.01 or P<0.05) . It also can improve the thymus index (P<0.01 or P<0.05) .Conclusion: Diacerein play an important role in the therapy of rheumatoid arthritis. The mechanism may be relevant to the role of reducing the levels of serum interleukin1-βand serum nitric oxide and the role of suppressing inflammatory reaction. It can also participate in regulating the immune system.
