This thesis reported a microchip-based capillary electrophoresis (MCE) electrochemical detection of tumor marker and DNA. We employed the system of orthoaminophenol (OAP)- H2O2-horseradis hperoxidase(HRP) to separate and detecte the tumor markers and DNA.In the first chapter, the current research situation of the microchip-based capillary electrophoresis (MCE) was described, and the electrochemical detection methods used in micrototal analysis was summarized.The second chapter describes the design and fabrication of the PDMS / glass composite chip and electrode.In the third chapter, the system of orthoaminophenol (OAP)-H2O2-horseradis hperoxidase (HRP) was investigated based on microchip-based capillary electrophoresis. Under optimum conditions, the linear range of free HRP is 4.5×10-9 7.8×10-13 M, the detection limit is 1.6×10-13 M.The forth chapter, a microchip-based capillary electrophoresis (MCE) electrochemical enzyme immunoassay protocol for the determination of carcinoembryonic antigen (CEA) and alpha-fetoprotein (AFP) was reported. Using this method, the linear range of AFP and HCG are 0.580.0 ng mL-1 and 1.7132.5 ng mL-1, respectively. The detection limit is 0.13 ng mL-1 and 0.30 ng mL-1, respectively.The fifth chapter establishes the detected method of 27-, 39-mer target DNA. With this protocol, the limits of quantification for the hybridization assay of 27-, 39-mer DNA fragments were of 8×10-12M and 1.2×10-11M, respectively. And we selected the detection of PCR amplifications from the Gene of E. coli to test the real applicability of our method. The proposed method has been applied satisfactorily in the analysis of E. coli genomic DNA.
