First of all, make the rats in allergic rhinitis group be allergic with ovalbumin(OVA)and enhance it and do local stimulation in order to establish the animal model of allergic rhinitis. While do intra-peritoneal injection with physiological saline in the same volume and intranasal dropping on the rats in normal control group.Rats in Montelukast group get the intranasal dropping on each side with OVA in the same volume, moreover, they given Montelukast(method:6mg/Kg. Bid). The levels of IL-25,IL-12 in serum of three groups of rats was examined with sandwich ELISA methods. The expressions of NF-κB and MMP-9 after immunohistochemical staining were observed. Compare the result with statistics software SPSS 13.0.Statistical analysis was performed using one-way ANOVA (difference between the two groups analyzed by t test), the linear regression and correlation. The data to mean±standard deviation indicated (x±S) by P-value<0.05 were considered as significant.Result: 1 The model on the rats in allergic rhinitis group is established successfully:all the rats in the allergic rhinitis groups show obvious symptoms:grasping noses,sneezing and running nose.2 The concentrations of IL-25 in three groups were Montelukast group: (32.8±4.6)pg/ml, allergic rhinitis group:(37.7±2.7)pg/ml. normal control group:(26.2±1.9)pg/ml respectively. The statistical test groups were compared:the difference between the three groups was significant significance (P<0.05).3 The concentrations of IL-12 in three groups were Montelukast group: (32.5±0.9)pg/ml,allergic rhinitis group:(26.1±0.6)pg/ml. normal control group:(36.2±1.4)pg/ml respectively. The statistical test groups were compared:the difference between the three groups was significant significance (P<0.05).4 The serum IL-25 level was negatively correlated with the serum IL-12 level in rats with Allergic rhinitis(r=-0.685,P<0.01).5 The remodeling of the nasal mucosa can be found in allergic rhinitis, cytoplasm and nucleal of epithelial and glandular cells displayed a more strongly positive staining reaction to NF-κB p65. The average optical of NF-κB p65(0.3613±0.0276) in group B were significantly higher than other two groups (P<0.001);The average optical of NF-κB p65(0.2366±0.0183) in group A were significantly lower than those in groupB,but higher than those in group C(0.1500±0.0225) (P<0.001).6 the cytoplasm of epithelial and lamina propria inflammatory cells a positive staining reaction to MMP-9. The level of MMP-9 in group B(0.2479±0.0254)were significantly higher than other two groups (P <0.001);The average optical of NF-κB p65(0.1795±0.0250) in group A were significantly lower than those in groupB,but higher than those in group C(0.1186±0.0252) (P<0.001).7 The level of NF-κB p65 was positively correlated with the The level of MMP-9 in nasal mucosa of rats with allergic rhinitis(r=0.879,P<0.01).Conclusion: 1 The animal model of the typical rhinitis symptoms and related to pathological changes in response (remodeling) changes can be established on the SD rats using the ovalbumin and enhance it and do local stimulation.2 The serum IL-25 level in rats with Allergic rhinitis was significantly increased, suggesting that IL-25 may involves in Allergic rhinitis and have the function of promoting inflammation development. The serum IL-12 level in rats with Allergic rhinitis was significantly lower than the control group, and There was a negatively correlation between the concentration of IL-25 and IL-12. The results indicate that imbalance of Th1/Th2 subsets with inhibition of Thl and predominance of Th2 exist in Allergic rhinitis subjects.3 The remodeling of the nasal mucosa can be found in allergic rhinitis. Growth factors such as NF-κB p65,MMP-9 express obviously in nasal mucosa in allergic rhinitis, The results indicats that NF-κB and MMP-9 might play an important role in nasal mucosa remodeling of rats with allergic rhinitis.4 To leukotriene receptor antagonists in the treatment group,The serum IL-25 level was significantly lower and was close to normal control group,and The serum IL-12 level was significantly increased, and was close to normal control group.
