| Objective:To observe the expression and distribution of Osteopontin(OPN) in Hypertrophic scar(HS) and Keloid (K),and expect finding new ways to diagonose and treat Pathological scar. Methods:The expression of OPN was examined by immunohistochemical analysis in serial sections of Pathological scar specimen obtained from 20 patients(10 of Hypertrophic scar group, HSG; 10 of Keloid group,KG) and 10 normal controls(10 of control group, CG).A part of common practice dehydrate, wrap to cover up, HE staining, immunohistochemical staining.The OPN expression as yellow or brown granules, use the Ax-70 to collect the sign and analysis by the ImagePro-Plus6.0(IPP6.0), measure each photograph in positive granules quantity, the average optical density(AOD) and the integral optical density(IOD).Statistical analysis was performed using GraphPad Prism 5.0, and all values were analyzed using the one-way ANOVA. Results:①Histomorphology:In CG, the stratum lucidum and the stratum corneum are thick; a lot of dermal papillaes stick up; a few fibroblast adhesion the collagenous fiber, small nucleus and acidophilia. In HSG, the stratum basale and stratum spinosum are thicker than in CG,but the stratum lucidum and the stratum corneum are lower;the dermal papillaes are short and reduced; the reticular layer is thick, collagenous fiber is hypertrophic and irregular; the number of fibroblast increased, volume larged, less basophilia. In KG, the stratum basale and stratum spinosum are thickest in three groups, but the stratum lucidum, stratum corneum and the stratum granulosum are atrophied; collagenous fiber is so irregularly; a lot of fibroblasts locate in, with big nucleus, less basophilia.②Immunohistochemical staining of the epidermis:The OPN expression as a yellow or brown granules.It expression in the stratum basale in CG and expression in the stratum basale and stratum spinosum in HSG and KG. Quantity of OPN positive granules among 3 groups:45.70±4.39,74.20±6.98,80.30±7.02. Compared with the CG,quantity of OPN positive granules in HSG and KG increased (P<0.05).The AODs among 3 groups:0.2246±0.0017, 0.2329±0.0010,0.2410±0.0018.Compared with the CG, AODs in HSG and KG increased. Compared with the HSG, AODs in KG increased(P< 0.05).The IODs among 3 groups:1293±45.52,1622±148.4,1924±134.3. Compared with the CG, IODs in HSG and KG increased(P<0.05).③Immunohistochemical staining of the dermis:The OPN expression as a yellow, yellow-brown or brown granules. It expressed significantly in dermis of HSG and KG,especially in KG Quantity of OPN positive granules among 3 groups:44.70±4.006,102.5±1.682,125.1±7.622. Compared with the CQ quantity of OPN positive granules in HSG and KG increased. Compared with the HSG, the indexes in KG increased (P <0.05).The AODs among 3 groups:0.1995±0.0021,0.2228±0.0017, 0.2329±0.0009.Compared with the CG,AODs in HSG and KG increased. Compared with the HSQ AODs in KG increased(P<0.05). The IODs among 3 groups:1092±21.65,1386±73.82,1968±76.49.Compared with the CQ IODs in HSG and KG increased. Compared with the HSQ AODs in KG increased(P<0.05).Conclusion:①OPN expression in normal skin and distribution mainly in the stratum basale.②OPN expression increased in pathological scar, especially in keloids.③The difference of the OPN expression and distribution mainly in dermis. |
