Objective:To study the expressions of Wnt-1 and Sfrp-1 in hypertrophic scar,keloid,surgical scar and normal skin tissues,and investigate their roles and significances in pathological scar.Methods:All these 48 patient scars(28 hypertrophic scars,8 keloids and 12 surgical scars)and 8 normal skin tissues were all originated from Department of Bums and Plastic Surgery,Xiangya Second Hospital,Central South University from March 2006 to March 2007(The recipients and donors all signed the informed consent.).They were stained by streptoavidin-biotion method of immunohistochemistry. Expressions of polyclonal antibody against Wnt-1 and Sfrp-1 and their correlation were determined.The scores of evaluation depended mainly on the straining degree and the positive cell counts of the sections.RESULTS:①The scores of Wnt-1 in the hypertrophic scar and the keloid were higher than those in the surgical scar and the normal skin (p<0.05).No significant difference was found between hypertrophic scar and the keloid,between surgical scar and the normal skin(p>0.05). ②The scores of Sfrp-1 in the surgical scar and the normal skin were higher than those in the hypertrophic scar and the keloid(p<0.05).No significant difference was found between hypertrophic scar and the keloid,between surgical scar and the normal skin(p>0.05).③The expression of Sfrp-1 was negatively correlated with wnt-1 in hypertrophic scar,keloid,surgical scar and normal skin.(r=-0.373,p<0.05).CONCLUSION:1 The overexpression of Wnt-1 in hypertrophic scar and keloid means the Wnt-1 may stimulate the formation of pathological scar.2 The overexpressions of Sfrp-1 in surgical scar and normal skin, which expression is low in hypertrophic scar and keloid,means the Sfrp-1 may inhibit the formation of pathological scar.3 Intervening the expression of Wnt-1 and Sfrp-1 may provide an effective method for clinical prevention and treatment of pathological scar.
