| Objective:To summarize the progress of the moden Chinese medicine research on COPD and the relationship between metabonomics and Chinese medicine. To study the exhaled breath condensate (EBC) of COPD patients with Qi deficiency syndrome and Yin deficiency syndrome with GC-MS based metabonomic measure.To explore the results of metabonomics and connotation of Modern science on COPD Qi deficiency syndrome and Yin deficiency syndrome, and to try to discover biomarkers.To provide a basis for the diagnosis and treatment of COPD with traditional Chinese and western medicines.Methods:The study included 17 Patients of COPD with Qi deficiency,15 Patients of COPD with Yi deficiency, and 16 healthy volunteers.After collecting the samples of EBC about 0.5ml, keep them in the refrigerator at -20℃.The EBS samples were analyzed with GC/MS, and the GC-MS results were analyzed by using the area normalization method, and each pear was determined. Based on the GC-MS results, hope to discover the small molecules which could differentiate the heealthy persons and the COPD patients with Qi deficiency and Yin deficiency.Results:1.Compared with the healthy volunteers, 1-[2-(Isobutyryloxy)-1-methylethyl]-2,2-dimethylpropyl 2-methylpropanoate in the Qi deficiency group increaseed markedly (20.69±14.05 vs 4.70±6.04,P<0.01),7,9-di-tert-butyl-1-oxaspiro(4,5)deca-6,9-diene-2,8-dione in the Qi deficiency group reduced significantly (0.48±0.34 vs 1.18±0.61,P<0.05),and 1-[2-(Isobutyryloxy)-1-1-methylethyl]-2,2-dimethylpropyl 2-methylpropanoate in the Yin deficiency group increaseed markedly(20.69±14.05 vs 4.70±6.04,P<0.01),phthalic acid, diisobutyl ester in the Yin deficiency group increaseed significantly(9.69±5.63 vs 3.81±4.77,P<0.05).2.Compared with the Qi deficiency, cedrol in the Yin deficiency group increaseed significantly(1.80±0.57 vs 0.96±0.61,P<0.05).Conclusion:1.Metabonomic study on exhaled breath condensate (EBC) by GC-MS could differentiate the healthy people and COPD patient, 1-[2-(Isobutyryloxy)-1-methylethyl]-2,2-dimethylpropyl 2-methylpropanoate,Phthalic acid 7,9-di-tert-butyl-1-oxaspiro(4,5)deca-6,9-diene-2,8-dione diisobutyl ester might be the potential biomarkers for COPD.2.GC-MS based metabonomic approach on EBC might be used for the differentiate the Qi deficiency or Yin deficiency, and cedrol might be a potential biomarker of Yin deficiency syndrome type.3.The EBC analysis showed a potential for disease diagnosis and study on Chinese medicine syndrome type. The current metabonomic method might be one of the useful measures to discover the evidence of Qi and Yin deficiencies and study on Chinese medicine syndrome type. |
