The Relationship And The Clinical Significance Between The Hp Infection And The Expression Of HIF-1α And Apoptosis-related Protein Bcl-2, Bax In Gastric Carcinoma | Posted on:2011-08-06 | Degree:Master | Type:Thesis | Country:China | Candidate:J Wu | Full Text:PDF | GTID:2154360308968156 | Subject:Internal Medicine | Abstract/Summary: | PDF Full Text Request | OBJECTIVE:To reveal the expression of HIF-1a,Bcl-2,Bax in gastric carcinoma tissues and the correlation of the three,and analyze the relationship between the expression of the three and Hp infection,as well as Hp infection,three kinds of factor expression and the clinical pathological features of gastric carcinoma, to explore the possible carcinogenic mechanisms of Hp infection,cell hypoxia and cell apoptosis.METHODS:Tissue samples were collected from 130 patients with gastric carcinoma and 70 patients with chronic gastritis, select the gastric carcinoma tissue samples as experimental group and the chronic gastritis tissue samples as control group. Hp was identified by rapid urease test and modified Giemsa-staining. Immunohistochemical method(PV9000) was used to detect the expression of HIF-1a,Bcl-2,Bax. The results were analysed for the relations among Hp infection, expression of HIF-1a,Bcl-2,Bax and their clinical pathological characteristics.RESULT:l.Hp infection:(1) Hp infection rate of gastric carcinoma group was significantly higher than control group (P<0.01).(2) Hp infection was correlated with depth of infiltration range and lymph node metastasis (P<0.05; P<0.05),but not related with cell differentiation grade (P>0.05).2. HIF-1a protein expression:(1) HIF-la protein expression of gastric carcinoma group was significantly higher than control group (P<0.01).(2) The expression of HIF-1a protein in gastric carcinoma tissue was correlated with depth of infiltration and lymph node metastasis (P<0.05; P <0.01),but not related with cell differentiation grade (P>0.05).3. Bcl-2 protein expression:(1) Bcl-2 protein expression of gastric carcinoma group was significantly higher than control group (P<0.01). (2) The expression of Bcl-2 protein in gastric carcinoma tissue was correlated with depth of infiltration and cell differentiation grade (P<0.01;P<0.01),but not related with lymph node metastasis (P>0.05).4. Bax protein expression:(1) Bax protein expression of gastric carcinoma group was significantly lower than control group (P<0.01). (2) The expression of Bax protein in gastric carcinoma tissue was correlated with depth of infiltration and cell differentiation grade (P<0.05; P<0.01),but not related with lymph node metastasis (P>0.05).5. The relations among the expression of HIF-1a,Bcl-2,Bax:(1) The expression of HIF-1a and Bcl-2 protein in gastric carcinoma tissue had positive correlation (P<0.01, r=0.515). (2) The expression of HIF-la and Bax protein in gastric carcinoma tissue had negative correlation (P<0.01, r=-0.510). (3)The expression of Bcl-2 and Bax protein in gastric carcinoma tissue had negative correlation (P<0.01, r=-0.578).6. The relations between the Hp infection in gastric carcinoma tissue and HIF-1a,Bcl-2,Bax protein expression and analysis (1)HIF-1a /Bcl-2 protein expression of Hp (+) group was significantly higher than Hp (-) group (P<0.05; P<0.01);Hp infection and the expression of HIF-la protein in gastric carcinoma tissue was positively correlated (P<0.05,r=0.255).(2) Hp infection and the expression of Bcl-2 protein in gastric carcinoma tissue was positively correlated (P<0.01, r=0.288).(3) Bax protein expression of Hp (+) group was significantly lower than Hp (-) group (P<0.01); Hp infection and the expression of Bax protein in gastric carcinoma tissue was negatively correlated (P<0.05, r=-0.536).CONCLUSION:1.Hp infection may be one of pathogenic factors of gastric carcinoma,it may promote invasion and metastasis of gastric carcinoma;Hp infection and the HIF-1a protein expression may play a synergistic role in the incidence of gastric carcinoma; Hp infection inhibits cell apoptosis, both were involved in the development and progression of gastric carcinoma.2.HIF-1a is closely related with the occurrence,the depth of invasion and lymph node metastasis of gastric carcinoma; Abnormal expression of Bax and Bcl-2 is involved in the development and progression of gastric carcinoma through the regulation of apoptosis and is closely related with depth of infiltration and degree of differentiation of gastric carcinoma.3.
