Obejective: To explore their roles of Livin and p73 in the development of cholesteatoma. Methods: Thirty cholesteatomas tissues and sixteen normal external ear canal skin were used in this study. All specimens were stained by immunohistochemical S-P method to detect the expression of Livin and p73, analysis was performed by the connection with clinical feature and the degree of ossicular destruction. Results: The expression of Livin was staining in the nuclei as well as the cytoplasm of cell and was expressed in full-thickness,mainly in the basal lamina of cholesteatoma epithelium.the staining of p73 were located in cell cytoplasm and was expressed in the spinous layer and granular layer.their positive rates were 76.7% and 40% in the cholesteatoma epithelium. The positive expression of Livin in the cholesteatoma epithelialium were increased greatly than that in external epithelialium(P<0.05),There was no significant difference between the expression of p73 in cholesteatoma and that in normal skin epithelial tissue(P>0.05).The expressions of above investigated indexes were not significantly different under the different degree of bone destruction and clinical feature(P>0.05). The expression of Livin in the middle ear cholesteatoma epithelium positively correlate with p73(r=0.411,P<0.05. Conclusions: The abnormal expressions of inhibitor of apoptosis protein Livin in the cholesteatoma epithelium suggests their important roles for in the formation and development of cholesteatoma, which may be involved in the regulation of apoptosis in the cholesteatoma epithelium.p73 maybe not play a role in middle ear cholesteatoma. The apoptosis and proliferation in cholesteatoma epithelium was not relate to the degree of ossicular destruction.
