| Objective: To analysis the structure characteristics of Bacteroides and Clostridia in intestinal microflora of normotension group and hypertension group in Kazaks, and to find out the structure differences in two groups. Methods:(1)We used 16SrDNA-PCR-DGGE to analyze the different structure of Bacteroides and Clostridia in intestinal microflora of these two groups in Kazaks. We defined the bacterial species through the cloning and sequencing of a different band and sequence alignment in Gene bank database. (2)We applied fluorescent quantitative 16S rDNA PCR to analyze and compare the bacterial relative quantity of Bacteroides and Clostridia in intestinal microflora between normotension group and hypertension group. Results: (1)Compared with these two groups, We found one dominant band from hypertension group .the relative frequency of this band had a statistically significant difference (P﹤0.05). We compared this obtained sequences with that of known bacteria listed in the Gene bank nucleotide sequence database by using Blast software. The bacterium was uncultured bacterium clone nbw1009b01c1. (2) Compared the logarithms of Bacteroides and Clostridia in intestinal microflora between normotension group and hypertension group, The number had significant differences ( P < 0. 01) . Conclusion: These findings support the deduction that the onset and development of hypertension in Kazaks may be in?uenced by both sides of the quantity of Bacteroides and Clostridia in intestinal microflora, and existence of uncultured bacterium clone nbw1009b01c1. |
