Objective: Using animal experiment, we research on the expansion law and histological change of Directed Skin Soft Tissue Expander (DSSTE), in order to prove DSSTE is harmless and available to the clinical patient.Methods: Using DSSTE and Routine Skin Soft Tissue Expander (RSSTE), we constructed the expanded flap models on miniature pigs in this randomized control trial. The research points of miniature pig which we embed expander contain cervix, scapular and haunch. During expansion interval, we use accurate body surface area measurement technology (Membrane Painting Method) to measure expanded flap in different volume of expander, protract area-volume scatter plot. We collect H.E, V.G and elastic fiber histological tissue cross sections to research histological changes in each group. Using digital image processing for medical applications (Image-Pro Plus 6.0) to measure the thickness of epidermis and the density of collagen fibers.Results: The expansion effect of DSSTE is significantly higher than RSSTE in the cervix. But in scapular and haunch, the expansion effect between DSSTE and RSSTE are not significantly different. Among the thickness of epidermis and the density of collagen fibers, both of them in expanded flap are significantly higher than normal condition. At the same time, the distributions in DSSTE and RSSTE groups are not significantly different.Conclusion: This research shows that only in cervix, DSSTE is effective than RSSTE. The histological changes between DSSTE and RSSTE are not significantly different.
