Objective: In recent years, many natural disasters such as snowstorms, earthquakes, tsunamis occurred frequently and took great pains and loss to the people of the countries and the regions. It has been an urgent task we faced that how to start prompt and effective treatments in these disasters. In the emergency treatment processes, acute infections, especially acute purulent infections and severe tissue damages always occur in disasters and can cause the increasement of neutrophils in blood. So testing the neutrophils quickly and accurately is significative for disease diagnosis and disease estimate. POCT is a strong developing testing technology in recent years, which means the analysis of the sample around the patients. Its main feature is simple and easy to use. Therefore, this issue starts from the neutrophils in blood reflecting the infections effectively. The ultimate goal is developing a portable, operating simply, rapid, accurate POCT product of detecting neutrophils without electricity and special areas.Methods:1. Separation of neutrophilsWe used two Percoll density gradient centrifugation methods, one with dextran, the other without dextran. Then we compared the advantages and the disadvantages of the two methods, and choosed the better one for getting the highly purified neutrophils in the follow-up experiments.2. Determination of enzyme activity in neutrophilsReferring to the neutrophil specific esterase staining method of bone marrow cells, and using urine dry chemical test strips for detecting urine neutrophil esterases, we selected one mothod that could reflect the number of blood neutrophils.Results: The Percoll density gradient centrifugation method without Dextran is simple and time-saving and can separate cells very well. The neutrophils we got had high purity and cell viability above 99%. The cells morphology was normal. Using urinary chemical test strips to detect blood neutrophils, we found that the color of the module changed, and the depth of color varied with the neutrophil concentrations. But the neutrophil specific esterase staining method of bone marrow cells can not be used for the determination of blood neutrophils.Conclusions:1. The Percoll density gradient centrifugation method without Dextran is simple and time-saving and can separate cells very well. The neutrophils we got had high purity and cell viability above 99%. The cell morphology was normal. It prepared for the neutrophils study in the follow-up experiments.2. Urinary chemical test strip can detect the neutrophils of blood samples, but only for the diluted blood samples of 0-500/μl neutrophils concentration. The neutrophils concentration of blood sample is much larger than the value above.Therefore, in follow-up experiments, we will continue to explore those substrates concentrations to direct detect blood neutrophils based on this method. |
