Objective:To detect the expression of hypoxia-inducible factor-1 alpha(HIF-1α) and vascular endothelial growth factor(VEGF) in hilar cholangiocarcinoma and investigate the correlation between two targets and their clinical significance,provide new reference for forecasting the carcinogenesis,invasion and metastasis of HC and judge prognosis.Methods:Specimens used for this study were obtained from 54 Hilar cholangiocarcinoma patients who underwent surgery from June 2004 to June 2007 and confirmed pathologically.ALL of the tissues were embedden in paraffin.There were 32 males and 22 females,aged from 39-78 years old(mean 61.6 years old).None of the patients had received radiotherapy,chemotherapy or immunotherapy before operation.Eleven cases of were used as normal controls.Immunohistochemical staining of HIF-1αand VEGF protein were detected with SABC method.Statiscal analysis was executed by Spssl4.0 software,P value less than 0.05 were considered significant.Results:(1) The positive expression rate of HIF-1αand VEGF in hilar cholangiocarcinoma tissues is 70.37%and 61.11%respectively.There was almost no expression of HIF-1αin nonhilar cholangiocarcinoma and VEGF expressed 9.09%in normal hilar bile duet tissues.There was significant difference between hilar cholangiocarcinoma and normal hilar bile duet tissues(P<0.05)(2) The positive expression rate of VEGF in hilar cholangiocarcinoma tissues witch HIF-1 expressed positively is 84.21%and the positive expression rate of VEGF in hilar cholangiocarcinoma tissues witch HIF-1αexpressed negatively is 6.25%.The expression of HIF-1αprotein had positive correlation with that of VEGF(rs= 0.542,P<0.05).(3) The expression of HIF-1αand VEGF in hilar cholangiocarcinoma tissues were both not significantly linked to gender,age,preoperative serum total bilirubin,Bismuth clinical type,tumor histopathologic grading(P>0.05) but both significantly linked to the capability of invation and transfer(P<0.05).Conclutions:(1) HIF-1αand VEGF were upregulated in hilar cholangiocarcinoma. There was significant difference between the hilar cholangiocarcinoma and normal hilar bile duet.This abnormality suggested that HIF-1αand VEGF should play a role in the tumorigenesis and progression of hilar cholangiocarcinoma.(2) The expression of HIF-1αand VEGF in hilar cholangiocarcinoma were significantly linked to the capability of invation and transer,which indicanting that HIF-1αand VEGF participated the process of invation and transfer in hilar cholangiocarcinoma.(3) The expression of HIF-1αprotein had positive correlation with that of VEGF in hilar cholangiocarcinoma.It is possible for them to make synergistic action during the tumorigenesis and progression of hilar cholangiocarcinoma. |
