Objective: We studies on HPLC fingerprints of the water- soluble constituents of different manufacturers of donkey-hide glue, provide experimental data scientific for establishing the quality standards.Methods: We used acetonitrilee and water as mobile phase with a gradient elution. The wavelength is 205nm.HPLC separation was performed at 25℃. The fingerprints of the water-soluble constituents in three kinds of different manufacturers of donkey-hide glue were gathered ;Simultaneously. we quantitated the phenylalanine in different donkey-hide glue by external standard method.Results: The results showed that alanine gelatin is higher than tyrosine in three different manufacturers. Two amino acids in Yang Gu donkey-hide were significantly lower than the other two manufacturers of donkey-hide.The gelatin alanine content in Dong E donkey-hide and Dong E Zhen donkey-hide is not much different. This show that the collagen of derm of different animal is different after the treatment. Different processes, the materials used in gelatin and animal skin origin may cause the different content of amino acids.Conclusion: Useing of extraction pretreatment, we studies on HPLC fingerprints of the water- soluble constituents of different manufacturers of donkey-hide glue. Establish the HPLC method for the amino acids of different manufacturers of donkey-hide glue. The research results provided evidence for establishing the quality of donkey-hide glue.
