Objective Hepatitis B virus(HBV) infection is an important global issue.China is a high HBV infection endemic areas of the world's by more than half.Human hepatitis B virus infection can lead to chronic asymptomatic HBsAg carrier(ASC),chronic hepatitis B(CHB),liver cirrhosis(LC)and hepatocellular carcinoma(HCC). Hepatitis B liver cell injury is caused mainly through a series of body immuse response.HLA,that is aware of the most complex genetic systems,plays an important role on antigen identification ,immune response and immune regulation,and is divided into three gene regions(ClassⅠ,ClassⅡand ClassⅢ) according to the different properties of their coding elements. ClassⅡmolecules associated with diseases are determined by its specific molecular structure.Polymorphism of ClassⅡis most frequent ,Polymorphism of HLA-DRB1 is most complex.People are trying to find susceptibility gene of hepatitis B to predict the determination of the development of diseases and guiding the treatment of hepatitis B.The researches on association between HLA-DRB1 and HBV infection have been launched in different areas and have made more progress,but the results in different parts of the crowd were not consistent.This study is to assess the association of human leucocyte antigen(HLA)-DBR1 gene with the outcome of hepatitis B in Tibetan of Qinghai.Methods 30 patients with cirrhosis due to HBV(B group)in Tibetan of Qinghai were investigated for HLA-DRB1 gene by polymerase chain reaction-sequence specific primers technique(PCR-SSP).The results were compared with those from 15 normal healthy people(A group) and 55 hepatitis B patients(C group)byχ2 test.We selected HLA-DRB1*03,*07,*12,*13,*15. This study is to assess the association of this five HLA-DBR1 gene with the outcome of hepatitis B in Tibetan of Qinghai.Results the frequency of HLA-DRB1*12 was the differences were of statistical significance (p﹥0.05),while the others were not significantly differen(tp<0.0125). With three group of the HLA-DRB1*12 binary comparison ,the frequency of HLA-DRB1*12 in B group was significantly higher (GF=90%,p<0.0125,RR=13.5) than the frequency of A group(6.7%);the frequency of HLA-DRB1*12 in C group was significantly higher (GF=98.2%,p < 0.0125,RR=14) than the frequency of A group(GF=6.7%);the frequency of HLA-DRB1*12 in B group was not significantly higher (GF=90%,p﹥0.0125) than the frequency of C group(GF=98.2%).Conclusion HLA-DRB1*12 may be the susceptibility gene in hepatitis B patients in Tibetan of Qinghai;and may be the susceptibility gene in the patients with cirrhosis due to HBV in Tibetan of Qinghai ; HLA-DRB1*03,*07, *13,*15 have no correlation with he outcome of hepatitis B in Tibetan of Qinghai.
