| Objective To investigate the expression of vascular endothelial growth factor(VEGF)in bladder transitional cell carcinoma(BTCC).Methods From January, 2008 to December, 2010, 60 examples paraffin wax specimen of bladder transitional cell carcinoma as a group were collected in first affiliated hospital of henan scientific and technical university.It included 38 males and 22 females.Aged from 45 to 75 years,the mean age was 58. The transitional cell carcinima was divided into third-level, namely high differentiation (Gl,n=24),middle differentiation (G2,n=21) and low differentiation (G3,n=15) .The urinary bladder carcinoma stages (UICC-TNM) were divided into two groups,including Ta~T1 (n=39) and T2~T4 (n=21) .And select the same period 10 cases of cases of normal bladder tissues as contrast group.10 cases of normal bladder mucosa from suprapubic prostatectomy surgical specimen.BTCC group and contrast group no significant age and gender differences.The expression of P27 and VEGF was detected by immunohistochemical technique in 60 cases of BTCC and ten cases of normal bladder tissue.And different grade and stage of bladder transitional cell carcinoma were compared and correlation analysis.Application SPPS17.0 statistical software for all data and chi-square test results, P<0.05 indicated significant difference, P<0.01 indicated very significant difference.Results (1)P27 protein was mainly expressed in the cell nucleus.BTCC group was 20% positive, positive expression in normal tissues was 80%,the difference was very significant (χ2=12.323,P<0.01 ) . P27 in bladder transitional cell carcinoma of the G1, G2, G3 expression rates were 37.5%, 9.5%, 6.7%,Among the three groups with low expression rate of the overall difference, there was significant(χ2=7.737,P <0.05,r=0.335);P27 in the expression of Ta~T1 group was about 41% in the expression of T2~T4 group was 14.3%, Ta~T1 expression rates slightly higher than the T2~T4 group, with statistical significance(χ2=4.51,P<0.05,r =0.274).(2)VEGF protein was mainly expressed in the tumor cell membrane, also expressed in the cytoplasm.It is positive in the BTCC group was 81.7%,The positive expression in normal tissues was 10%,Significant difference (χ2=18.203, P<0.05) . VEGF in bladder transitional cell carcinoma of the G1, G2, G3 expression rates were 75%, 88.2%, 92.7%, among the three groups with high expression rate of the overall difference, there was no statistically significant (χ2 =2.379, P>0.05) .VEGF in the Ta~T1 group, the expression rate was64.1%, in T2~T4 group, the expression rate of 90.5%, T2~T4 group expressed higher than that in Ta~T1 group, with statistical significance (χ2=4.855, P< 0.05, r =-0.284) .Conclusion (1)P27 in bladder transitional cell carcinoma significantly lower than normal bladder tissue, both compared statistical significance (P<0.01) .(2)VEGF in bladder transitional cell carcinoma was significantly higher than the normal bladder tissue, compared to statistical significance between the two (P <0.01) .(3)P27 in bladder transitional cell carcinoma and clinical of the pathological factors prompted with tumor grade and stage increase, P27 expression is significantly reduced.(4)VEGF expression and clinical pathological factors of the tips with the tumor grade and stage increased, VEGF expression was significantly increased.(5) Bladder transitional cell carcinoma of the expression level of P27 was significantly reduced and the expression levels of VEGF were significantly increased, may be associated with bladder transitional cell carcinoma, development, metastasis,comprehensive testing will help determine the expression of both the biological behavior of BTCC. |
