Association Analyses Of SUR1 Gene And KCNJ11 Gene Polymorphisms With Gestational Diabetes Mellitus In Guang Dong Hans In China | Posted on:2011-04-30 | Degree:Master | Type:Thesis | Country:China | Candidate:Q Feng | Full Text:PDF | GTID:2154330338476830 | Subject:Biochemistry and Molecular Biology | Abstract/Summary: | PDF Full Text Request | ObjectiveGestational Diabates Mellitus (GDM)is a metabolic disease with complex genetic and environmental components.It has many genetic and environmental components similar to type 2 diabetes.We investigate the allelic frequency of Sulfonylurea receptor 1 gene ( SUR1 ) and Kalium inwardly recitifier gene (KCNJ11)of gestational diabetes mellitus (GDM) women to find out the susceptible gene associated with GDM and the relationship between these genes and the clinical characteristics.MethodsFifty-three GDM women were served as study group, fifty normal pregnant women were selected as control group.The distribution of SUR1 rs1799854,rs1799859 and KCNJ11 rs5219 alleles frequencies of these pregnant women were examed by polymerase chain reaction(PCR)-restriction fragment length polymorphism (RFLP) method combined with direct sequencing. Height,weight,blood pressure and fasting blood glucose were measuered in all subjects.Independent sample t test was used to compare means between two groups.ANOVA was employed to compare the differences of clinical characteristics between genotypes.Chi-square test was employed to test the the differences of genotypic and allelic frequencies between cases and controls.Association analyses were done by logistic regression.All statistical analyses were calculated by the software of SPSS 12.0 for windows. Results:1.There was significantly difference of the frequency of genotype GA (rs1799859) between GDM group and control group (32.1% vs 8.0%,p=0.002). Also, there was significantly increased alleles frequency of A(rs1799859) between GDM group and control group. (16% vs 4%,p=0.004).2.There was significantly difference of the frequency of genotype EK,KK(rs5219) between GDM group and control group (71.1% vs 14.0%,20.8% vs 14.0%,p=0.038).3. Results from multiple logistic regression analysis showed that the A - G transition polymorphism of SUR1 rs1799859 (OR = 6.82,95 %CI 1.90~24.54) and the A - G transition polymorphism of KCNJ11 rs5219 ( OR = 3.90, 95 %CI 1.02~15.00) were associated with the development of GDM.4.The association between SUR1 rs1799854 and the development of GDM was not found.Conclusions:The SUR1 gene 31 exon polymorphism (rs1799859) and the KCNJ11 gene E23K polymorphism (rs5219) are associated with GDM in Han Chinese in Guang Dong. These polymorphism may have a diabetogenic effect by changing the function of ATP-sensitive KATP channels, which impair glucose-induced insulin release of the health and make pancreaticβ-cell oversensitively.
