AIM:TO study the effect of berberine on the growth and invasion and metastasis of human gastric carcinoma cell line MGC-803.METHODS:Cells were divided into the control group(MGC-803 cells only)and the berberine-treated group ( Corresponding concentration berberine ) The measurement of inhibitory rate of berberine were done by MTT assay.The positive expression rate of ICAM-1,MMP-2,CD147,iNOS,Survivin and TIMP-2 were detected by immunocytochemical staining and Western blotting. The expression of ICAM-1,MMP-2,CD147,iNOS,Survivin and TIMP-2 mRNA were observed by Real-time quantitative reverse transcription polymerase chain reaction(qPCR)。RESULTS:Berberine could inhibit the growth of gastric carcinoma cells obviously in a dose and time-dependent manner from 0μmol/L to 40μmol/L.Experiments of immunocytochem-. ical staining and Western blotting show that a great deal of expression of ICAM-1,MMP-2,CD147,iNOS,Survivin and TIMP-2 protein can be detected in the gastric cancer cells group, but after 18h since the disposal of the berberin, the expressions of ICAM-1 ,MMP-2, CD147, Inos, Survivin protein decreased significantly (P <0.01) and TIMP-2 protein increased significantly (P <0.01). The comparation between the two experiments have the statistics signification(P<0.01).It show in the experiment of Real-time RT-PCR of ICAM-1,MMP-2,CD147,iNOS,Survivin and TIMP-2 mRNA in gastric cancer cells, that the expression of ICAM-1,MMP-2,CD147,iNOS,Survivin mRNA decreased and TIMP-2 mRNA increased significantly after 18h since the dispos- al of the Berberine, and the comparation between the two experiments have the statistics signify cation(P <0.01).CONCLUSION:Berberine can inhibit the growth and invasion metastasis of human gastric carcinoma cell line MGC-803 in a dose and time-dependent manner.Its mechanisms may be relevant to the regulation of ICAM-1,MMP-2,CD147,iNOS,Survivin and TIMP-2 protein expression and control ICAM-1,MMP-2,CD147,iNOS,Survivin and TIMP-2 mRNA.
