| Objective:Studies to investigate these changes of the tumor necrosis factor-α(TNF-α), interleukin-6 (IL-6) in blood serum , the macrophage inflammatory protein-1α(MIP-1α)in lung homogenate,the immunohistoch-emical method on the lungs organization and the structures of lungs organization under the electron microscope is for the purpose of discussing lungs' protective function ofω-3 polyunsaturated fatty acids for rat injuried thermally.Method:72 SD adult health rats were randomly divided into three groups : normal group(n=8), the B group ,short for 20% long chain fatty acid treatment group (n=32) ,the C group ,short forω-3 polyunsaturated fatty acids treatment group (n=32). The B group and C group rats use 30% total body surface area(TBSA) full thickness burns model and the two group of rats was injected medicine by tail vein .Every group of rats were divided into four scheduled time points (1d, 4d, 7d, 10d, ) to check these changes of the tumor necrosis factor-α(TNF-α), interleukin-6 (IL-6) in blood serum and the macrophage inflammatory protein - 1α(MIP-1α) in lung homogenate.The immunohistochemical method on the lungs organization to check the level of NF-κBp65 and MIF.Research the structures of lungs organization under the electron microscope.Results:The levels of TNF-αand IL-6 in the blood serum of B group and C group is upper than the normal group (P<0.01) when the rats were burned seriously in the first day ,then gradually be upper ,it were be at the peak in the seventh day .Then, gradually be lower but it still was upper than that of A group (P<0.01);The change trend of the level of MIP-1αof the lung organization slurry was the same as that of the TNF- αand IL-6 in the blood serum, It was the highest level in the seventh day ,and then gradually be lower .To compare with B group ,the levels of TNF-αand IL-6 in the blood serum and MIP-1αof the lung organization slurry of C group were obviously lower (P<0.05) .The expression of NF-κBp65 and MIF of C group is lower than that of B group .The damage of The lung steepsⅡtype cell of C group is less than B group .Conclusion:(1)Theω-3 PUFAs effectively lowers the contents of TNF-α,IL-6 in rat's blood serum. (2) Theω-3 PUFAs effectively lowers MIP-1αcontents of rat's lung organization slurry.(3) Theω-3 PUFAs effectively reduces the expression of NF-κBp65 and MIF by immunohistochemical method on lung origination .(4) The anti-inflammatory mechanism of theω-3 can block the activating of signal path of NF-κB to reduce a variety of inflammation mediators and protect the rats'lung when the rats were be burned seriously . |
