0bjective To investigate the effects of angiotensin II type 1 receptor and gene in NAFLD pathogenesis.Methods After one week's adaptive feeding,32 rats were randomly divided into control group (ordinary diet)and model group(high-fat diet)equally,8 rats were killed at the every tenth week,fourteenth week in each groups,liver function and blood fat were monitored by automatic biochemistry analyzer, angiotensin ( Aâ…¡) and aldosterone(ALD)by radioimmunoassay counter,the expression of AT1R in liver tissue by immunohistochemistry,the extraction of male rats RNA in liver tissue by Trizol,the expression of AT1R in liver tissue by reverse transcription polymerase chain reaction(RT-PCR).Results 1,Body weight in model group was higher than the normal control group at the end of tenth week,while it was lower than the normal control group at the end of fourteenth week;2,Liver weight,serum ALT,TC,TG,LDL-C and angiotensin(Aâ… ),Aâ…¡were all higher than those of normal control group,while the HDL-C was significantly lower at the end of tenth and fourteenth week;3,Serum angiotensin 1,2 were higher than those of control group, and there was no statistical significance of Aâ… (P>0.05)10 weeks later;4,Serum angiotensin 1,2 were higher than those of normal control group, and there was statistical significance by the end of 14 weeks;Serum ALD was all lower than normal control group with no statistical significance;5,Epression of AT1R existed in model group,with higher expression level at the 14th week,there was nearly no expression in control group;6,two groups all could extraction of RNA,model group higher expression AT1R mRNA than control group.Conclusion Angiotensin II type 1 receptor and gene were expressed in nonalcoholic fatty live rats, the expression level rise gradually with the aggravation of the fatty liver lever.
