| Objective To study the protective effect of Panax Notoginseng Saponin in the hypoxic-ischemic myocardial injury through establishing the classic neonatal rat model with neonatal rat atmospheric Asphyxia model. Through testing the changes of CK-MB and cTn I in serum, comparetion the characteristics of CK-MB and cTn I. Study the sensitivity and specificity of CK-MB and cTn I in myocardial injury, to verify the protective effect of Panax Notoginseng Saponin in hypoxic-ischemic myocardial injury. To seek a new theoretical foundation for the treatment and diagnosis of hypoxic-ischemic myocar-dial injury in neonatal asphyxia.Methods 88 7-day-old newborn SD rats were randomly divided into the control group, saline-treated group and PNS-treated group. Right after hypoxic-ischemic insult, PNS or normal saline solution were injected intraperitoneally. Normal Saline-treated group or PNS-treated group were also randomly divided into saline or PNS-treated for 6 hours(group 6h),24 hours(group 24h),48 hours(group 48h),72 hours(group 72h),7 days(group 7d). The newborn rats of PNS-treated group were given 100mg/kg PNS by intraperitoneal injection as soon as the neonatal rat model with neonatal rat atmospheric Asphyxia model were established, the same dose of PNS was repeated by intraperitoneal injection after 12 hours. The newborn rats of Normal Saline-treated group were given the same volume of normal saline by intraperitoneal injection.At the end of the study, the level of CK-MB and cTnl in plasma was measured. Heart tissues were also harvested for measurement using HE. At last the data were processed with SPSS 13.0 statistical method.Results:(1) Compared with the control group, the level of CK-MB and cTnâ… was time-dependently increased in saline-treated group between 6 hours and 24 hours, while treated for 3 days and 7days, the level of CK-MB was decreased and normal. However, the level of cTnâ… was still higher than normal when treated for 7days. (2) Compared with the saline-treated group, the level of CK-MB and cTnâ… was decreased in PNS-treated group at any time. Meanwhile, the peaks of the level of CK-MB and cTnâ… were also lower in the PNS-treated group. However, the level of cTnâ… was normal in PNS-treated group when treated for 7 days. (3) Compared with the control group, the level of CK-MB and cTnâ… was increased in PNS-treated group, and the level of CK-MB and cTnâ… reached the peak when treated for 24hours,while the peaks were lower than saline-treated group. The level of cTnâ… was normal when treated for 7 days.Conclusions:These findings suggest thatâ‘ PNS can attenuate cardiac injury induced by myocardial ischemia and anoxia.â‘¡CK-MB and cTn I play important roles in evaluating and detecting the cardiac injury. |
