| Fatigue is kind of subjective discomfort, but objectively, people who is suffering fatigue will lose their normal working and living abilities. Normally, when organism is strongly stimulated by external substance or stimulated for a long time, abilities of function or action of cells, tissues and organism will reduced, such as fatigue of muscle cells and tissues. Fatigue and hypoxia always come one after another. A lot of oxygen will be used during strong exercise. When oxygen can not be supplied very well, organs will suffer hypoxia. In another condition, when organs are suffering hypoxia, anaerobic metabolism will play the key role, followed by metabolic waste such aslactic acid which will make microenvironment changed. At the end fatigue will be come. Decrease of action ability of muscles is an essential feature of fatigue. What we are focus on is seed of Hovenia dulcis thunb, which is a kind of plant belong to Hovenia Thunb genus Rhamn-aceae family. Hovenia dulcis thunb has profit for protecting liver, diuretic, blood pressure, pain, CNS depressed, anti-tumor and antioxgen. Extract of Hovenia dulcis thunb via water extraction and alcohol precipitation-grind fine seeds of Hovenia dulcis thumb, and then use 4 times water to reflux 2h for each times total 3 times, and then filter and concentrate it. Finally, add ethanol which final concentration is 50%, and then store it in fridge for 48h. Filter and dry it which was just what we want-was confirmed before that it had anti-fatigue and anti-hypoxia activities. In this paper Ethyl acetate extract which contains more flavone is obtained via a series of extraction steps, and compared with extract of Hovenia dulcis thunb via water extraction and alcohol precipitation using treadmill model of rats and hypoxia model of cells. As a result, the extract process was obtained that extract of Hovenia dulcis thunb via water extraction and alcohol precipitation was dissolved by eight times water and heated 30min, and then the upper liquid was extract via ethyl acetate (1:2,v:v) three times, and then concentrate ethyl acetate part. Through detection of blood glucose, liver hepatic glycogen, muscle glycogen, acetylcholinesterase, lactate dehydrogenase, lactic acid, urea nitrogen, malondialdehyde, superoxide dismutase and total antioxidant activity, extract via water extraction and alcohol precipitation and ethyl acetate extract of Hovenia dulcis thumb were confirmed that both of them had anti-fatigue activity. Extracts of Hovenia dulcis thunb maybe mainly affect energy metabolism, free radical metabolism and enzymes to resistance fatigue. Cell biological researches were also mentioned in this paper. We have used special incubator to keep cell without oxygen for 24h, and then detected cell viabilities. The results confirmed again that extracts of Hovenia dulcis thunb were effective on fatigue. The same effects of quercetin and dihydromyricetin were also proved. Considering all the results flavone was supposed to be the main effective substance. |
