It is presently considered that the incidence of gliomas is closely related with the gene mutation, related genes research has become a hot spot on diagnosis and treatment of glioma. Research has shown that SOX-2 genes and TBX2 genes involved in the occurrence and development of glioma by multiple mechanisms. SOX-2 may regulate the differentiation of brain tumor stem cells to participate in the incidence of gliomas. TBX2 may inhibit tumor cell apoptosis through the negative regulation of multiple tumor suppressor genes. There is no reports about the expression of SOX-2 and TBX2 in glioma. In this study, the protein and nucleic acid of SOX-2 and TBX2 expression in glioma were detected by immunohistochemistry and RT-PCR to evaluate their clinical significance.Methods:59 specimens of glioma surgery (from part 2 of First Hospital of Jilin University, and Third Hospital of Jilin University, March 2009 - June 2010), in which 6 cases are gradeâ… , 21 cases are grade II (I-II grade are a total 27 cases of low-grade gliomas),24 cases are grade III, 8 cases are grade IV (III-IV grade are a total 32 cases of high-grade gliomas). In addition, there are 9 cases of normal brain tissue. The expression of SOX-2 and TBX2 in normal brain tissue, low-grade gliomas and high-grade gliomas were dected by immunohistochemistry and RT-PCR. Statistical Analysis adopt SPSS ll.5 software package.Results:With the detection of immunohistochemistry SOX-2 and TBX2 protein provided positive results.And it was consistent with RT-PCR.SOX-2 has a high positive expression in gliomas which is significantly different with the expression in normal brain tissue (P<0.05).
