Objective:To study the expression of Tbx2,Tbx3 in Primary Hepatocellular carcinoma cells and normal hepatic cells. To analyze the change of Tbx2 and Tbx3 in the development of HCC.Methods:(1) Thirty-two specimens of Primary Hepatocellular carcinoma and non-cancerous adjacent liver tissues were collected and then fixed in formaldehyde solution routinely. After embedded in paraffins, the specimens were cutted into slices and dyed by hematoxylin-eosin staining to differentiate the nature of cells under the microscope. The slices of hepatocellular carcinoma tissue were detected by immunohistochemistry.(2) Immunohistochemistry kits:Goat anti-human Tbx2/Tbx3 polyclonal antibody(from Santra Cruz),SP immunohistochemistry kits(from Zhongshan Biotechnology Corporation,Beijing).(3) Slices were sealed by neutral gum after immunohistochemistry,and observed under the microscope.Then effective areas were photographed.(4) It was regarded as positive that endochylema and karyon were dyed. Semiquantitative analysis were took according to amount of positive cells and dyed intensity:no positive cells, scored 0;1%-30% were positive, scored 1; 31% -70% were positive, scored 2; 71%-100% were positive, scored 3. Then slices were also scored according to dyed intensity(non-staining slices,scored 0; light brown ones, scored 1; claybank ones, scored 2; sepia ones, scored 3).two scores of every sliceds were summed.0 score was regarded negative; 1-2 scores were regarded as weak positive(+);3-4 scores were regarded as positive(++);5-6 scores were regarded as strong positive(+++). (5) Specimens of breast cancer, colorectal cancer, pancreatic cancer and normal pancreatic tissue were collected and detected by immunohistochemistry by the same method, comparing with related literatures.Results:(1) The expression of Tbx2 and Tbx3 were both observed in hepatocellular carcinoma cells and normal hepatic cells.(2) The strong positive rate of Tbx2 was 0% in hepatocellular carcinoma cells, while it was 87.5% in normal hepatic cells. The data were analyzed by x2 analysis. There was significant difference compared the expression of Tbx2 in hepatocellular carcinoma cells and that in normal hepatic cells(P<0.01).(3) The strong positive rate of Tbx2 was 0% in hepatocellular carcinoma cells, while it was 90.6% in normal hepatic cells. The data were analyzed by x2 analysis. There was significant difference compared the expression of Tbx3 in hepatocellular carcinoma cells and that in normal hepatic cells(P<0.01).(4) The expression of Tbx2 and Tbx3 in hepatocellular carcinoma cells and normal hepatic cells were both weak positive with the former was weaker than the latter.there was no significance in all differentiation types.(5) The results of breast cancer, colorectal cancer, pancreatic cancer and normal pancreatic tissue showed no difference with related literatures.Conclusions:(1) The expression of Tbx2. Tbx3 changed in the cancerization process of hepatic cells.(2) Tbx2. Tbx3 were immortalization genes, which were consanguineously related to the tumors'occurring and development. Whether the occurring of tumors is related to mutation of genes or anomalism of conduction path. Further studies are needed.(3) Mechanism of hepatocellular carcinoma is different from breast cancer, colorectal cancer and pancreatic cancer. There is significance in molecular level.
