Objective:To fabricate an absorbable membrane for guided bone regeneration in dental implant with chitosan and collagen,test and assess the physical property of this membrane.Methods:2% chitosan acetic acid solution and 0.3% collagen aqueous solution were mixed to be chitosan-collagen solution of Group A, B, C in the volume proportion of 2:1, 1:1, 1:2. Then the mixed solution were frozen and dried to be membranes. Modulus of elasticity, yield strength, breaking strength, elongation at break of the membranes were tested through elongation test, the results were compared and analyzed by t test. Porosity and pore diameter(area) of the membranes were tested by mercury prosimeter. Surface structure of chitosan-collagen membrane and Bio-Gide membrane were observed by scanning electron microscope and compared. The physical property of chitosan-collagen membrane was assessed.Results:(1) 2% chitosan acetic acid solution and 0.3% collagen aqueous solution can be mixed uniformly. The mixed solution can be frozen and dried to be membranes. The chitosan-collagen membranes of Group A, B, C were fabricated in the volume proportion of 2:1,1:1,1:2 successfully. Membrane thickness of Group A was about 0.30 mm-0.50 mm, the texture was uniform, the strength and toughness was good. Membrane thickness of Group B was about 0.30mm-0.50mm, the texture was relatively uniform, the strength and toughness was fine. Membrane thickness of Group C was not equal, the texture was not uniform, the strength and toughness was poor. (2) Modulus of elasticity, yield strength and breaking strength of the membranes of Group A were significantly better than Group B (p<0.01). Elongation at break of the membranes of Group A and Group B have no significant difference (p>0.05). (3) Porosity of the membrane of Group A was 90.95%, median pore diameter (area) was 15.6 um; porosity of Group B was 86.96%, median pore diameter (area) was 13.9 um. (4) Scanning electron microscope observation showed the surface of chitosan-collagen membrane of Group A was generally even, but it had bumps and holes, the membrane was polyporous, the diameter of bigger pore was about 20 um while the smaller was about 7 um; the surface of Bio-Gide membrane was generally even, but it had bumps and holes, the diameter of most pores were less than 1 um.Conclusion:2% chitosan acetic acid solution and 0.3% collagen aqueous solution can be mixed uniformly to be membrane by frozen and dried. The membrane fabricated in the volume proportion of 2:1 had proper physical property,making it an ideal material of absorbable membrane for guided bone regeneration in dental implant.
