Objective: To investigate the role of connexin 40 of trigeminal ganglion in migraine attack and the change of periorbital skin pain threshold.Methods: Healthy adult SD rats were randomly divided into sham surgery group, control group, experimental group which according to different investigate time following the stimulation of rat dual matter adjacent to the superior sagittal sinus to divided into four subgroup. The change of expression of connexin 40 of each group using immunofluorescence technique and western blot analysis was observed. The pain threshold of periorbital skin using von frey hair to monitor was evaluated.Results: Immunofluorescence showed that the connexin 40 expression of experimental 0h group, experimental 12h group, experimental 24h group were extremely higher than sham surgery group,control group, experimental 48h group(p <0.05), the top time point was at 12h, in each group the expression was equal(p>0.05). Western blot analysis showed that the connexin 40 expression of experimental 0h group, experimental 12h group, experimental 24h group were very intensity higher than sham surgery group, control group, experimental 48h group(p <0.05), the top time point was 12h, in each group the expression was equal(p>0.05). Von Frey hairs showed that the periorbital skin pain threshold of experimental before 1h group, experimental 2h group, experimental 12h group, experimental 24h group, experimental 48h group at first decreased and than increasd, the lowest timepoint was experimental 12h group, experimental 48h group returned to normal level.Conclusions: We found that following the stimulation of rat dual matter adjacent to the superior sagittal sinus expression of connexin 40 increased, pain threshold of periorbital skin decreased and periorbital skin sensitization increased. We proposed that connexin 40 may contribute to migraine.
