Objectives:There is few study on therapy of central sensitization in patients with migraine. Pregabalin has good prospects on treating central sensitization but less been used in the treatment of migraine. The predictors of allodynia in migraine and the effects of pregabalin in alleviating allodynia were investigated in this study.Methods:Sixty-three consecutive patients with migraine diagnosed according to International Headache Society criteria were prospectively investigated. Detailed information of the patient’s course of disease, severity and frequency of migraine and basic personal information were recorded. The cutaneous allodynia symptoms that occurred during headache attacks were documented using an Allodynia Symptom Checklist of Chinese version for assessing cutaneous allodynia and the predictors of allodynia were analyed. Fourty-one patients with allodynia were treated with pregabalin (150-300mg/d) for12weeks. The improvements in allodynia and headache severity were recorded at1and3months. The side effects of the drugs were also recorded.Results:The average age of all patients was36.3±13.0years, and66.7%were females.65.1%patients had allodynia:thermal in85.4%, static mechanical in29.4%, and dynamic in22.0%. The allodynia appears to be associated with female gender, duration of illness, headache attack, longer duration of headache, higher values of MIDAS and medication-overuse.Compared with baseline, the migraineus with CA reduced from41cases to31cases after3-months treatment. The ASC score decreased from mild or moderate CA to mild CA. Both scores of ASC and three kinds of patients of CA decreased significantly (P<0.05). The frequency and severity of headach, and MIDAS、HIT-6score were significantly decreased (P<0.05) when compared with baseline. Drug efficiency reached85.4%. The composite side effects were mild and tolerance.Conclusions:1. There were41migraineurs had allodynia screened through the ASC questionnaire and the incidence rate was65.1%. These patients need more attention and standard treatment since the high incidence of allodynia.2. The allodynia appears to be associated with female gender, duration of illness, headache attack, longer duration of headache, higher values of MIDAS and medication-overuse. The relationship between allogynia and aura, age. symptoms accompanied with migraine were not defined yet.3. Accroding to the ASC. allogynia of thermal was most common (85.4%), and the static mechanical in29.4%, and dynamic in22.0%.4. Pregabalin can not only improve the quality of life, reduce the attack, the duration and severity of headache also reduce the attack of allodynia in patients with migraine. Pregabalin was effective in relieving allodynia.
