The Study Of Expression's Meaning About Stem Cell Gene Oct4 In Gastric Cancer Cells

Objective:To investigate the significance and expression of stem cell gene Oct4 by semi-quantitative RT-PCR in three gastric cancer cells MKN28,SGC7901 and BGC823,and then,to interfer the expression of Oct4 in gastric cancer cell which has a highest exprssion degree of Oct4 by using a technique called RNA interference.After the Oct4 interfered,to investigate the invasive changes of gastric tumor cells,and then to explore the relationship between Oct4 gene and the malignant degree of gastric tumor cells.Methods:1. to detect Oct4 mRNA expression from high malignant degree to low among the three gastric cancer cells lines by semi-quantitative RT-PCR technology,and then, to find the gastric cancer cells line with high expression degree of Oct4 .2.to interfer the expression of Oct4 with Oct4 siRNA in the gastric cancer cells with high Oct4 expression degree,and then to detect the expression degree of Oct4 by the fluorescent quantitative polymerase chain reaction and Wstern-bloting technologies after interference.3.to compare the changes of gastric cell invasion among the non-dealing,negative siRNA and Oct4 siRNA ganstric cancer cell by Transwell cell invasive trial in vitro.the same experiment was repeated three times.Result:1.Semi-quantitative RT-PCR result indicates that the Oct4 expression degree from high to low in turn is BGC823,SGC-7901,and MKN28 in the three gastric cancer cell lines .2.After interfering the Oct4 in BGC823 cells,the results of fluorescence quantitative PCR and Western-bloting show that the Oct4 exprssion degree decrease evidently between non-dealing and Oct4 siRNA interference,and the differnces are all statistically significant(p<0.05). 3.The results of Transwell cell invasive trial in vitro indicate that the cell mount of Oct4 siRNA interference through membrane is significantly declined compared with the non-dealing and negative siRNA cell respectively,and the differences are statistically significant(p<0.05).Conclution:1.The differentiation degree of three gastric cancer cell lines from low to high is BGC823,SGC-7901 and MKN28 in turn,at the same time ,the degree of Oct4 expression from high to low was also BGC823,SGC-7901 and MkN28 in turn,and this phenomena suggests that Oct4 may play an important role in maintaining the tumor poorly differentiated state.2.The invasive ability of BGC823 is dropped significantly after Oct4 interference,and this phenomena suggests that there may be an direct correlation between the Oct4 expression and the malignant invasive ability of gastric cancer cells,and then hints that Oct4 may play an important role in the invasive aspect of gastric cancer cells.
