Objective:This paper explores the influence of different concentrations of dandelion on the proliferation, migration, invasion and expression of relative protein of H22 cell of Hepatoma and BGC823 cell culture in vitro of human gastric cancer, and makes preliminary discussion on the anti-cancer function and molecular mechanism of dandelion.Methods:Applying IHC to detect the influence of different concentration of dandelion drugs on the expression standard of the TGF-β1,mTOR, MMP-2 and MMP-9 Cytokine-Protein. Applying clone formation and MTS to detect the influence of different concentration of dandelion on the cell survival rate and change of proliferation of BGC823 applying Transwell Chambers to detect the influence of different concentration of dandelion on the cell migration and invasion of BGC823 cell; applying real-time quantitative PCR method to detect influence on the expression standard of MMP-2mRNA of BGC823 by different concentration of dandelion drugs.Results:1.The result of in vivo shows that the different concentration of dandelion drugs inhibit the proliferation of H22 cells of Hepatoma, compared with the control group, the difference is significant(P< 0.05).The result of IHC shows that dandelion drugs down-regulate the expression quantity of TGF-β、mTOR、MMP-2 and MMP-9 of the cancer cells.2.All the three different concentrations of dandelion drugs inhibit proliferation of BGC823 cell of gastric cancer, and with the increase of the concentration, the quantity of clone cells decreases significantly (t=2.000,1.732,1.527, P<0.05);2.5mg/ml dandelion inhibits the proliferation of BGC823, and as the culture time extends the increase of cells slow down; 4.0mg/ml dandelion has the strongest inhibition on the proliferation of BGC823 (P<0.05)3. The result of Transwell Chambers test shows that all the three different concentration of dandelion decrease the quantity of the migration ofBGC823, and with the increase of the concentration the effect becomes stronger. Compared with the control group, the 2.5 mg/ml and 4.0mg/ml group have significant difference with it (t=0.359、0.297, P<0.05), but there is no big difference between the two group (P>0.05), it shows that in a certain extent of concentration the change of concentration of dandelion makes small change on the migration of cancer cells. Dandelion basically has the same influence on the invasionBGC823 with migration.4. According to the results of RT-PCR, the different concentration of the dandelion drugs solution all down-regulate MMP-2RNA expression of BGC823, and the effect of down-regulation increases with the increase of the concentration; when concentration is 2.5 mg/ml and 4.0mg/ml, there is significant difference on the down-regulation of expression of MMP-2 (P<0.05)Conclusion:1. Dandelion inhibits the growth of the H22 cells of Mouse Hepatoma,0.25g/ml has the significant inhibition to the growth of the H22 cells of Mouse Hepatoma.2. Dandelion down-regulates the expression of TGF-β1 and mTOR, so it inhibits the expression of the relative genes in the middle and lower reaches of signal pathway, and it inhibits proliferation of cancer cells. Dandelion also down-regulates the MMP-2 and MMP-9 which are the two main molecular controlling the movement of cells, it proves that dandelion inhibits the change of expression of relative genes to inhibit the proliferation and migration of cancer cells.3. Dandelion has a significant inhibition effects to BGC823 culture in vitro, and it probably functions through down-regulating the expression of MMP-2 and MMP-9 in BGC823 to inhibit the migration and invasion of cancer cells.
