| Objective:In clinical practice,the incidence of thyroid cancer is higher among Hashimoto’s thyroiditis(HT),especially for papillary thyroid carcinoma(PTC).BRCA1 and 53BP1 are DNA damage response proteins which play an important role in maitaining stability while the expression of them in thyroid tissue is unclear and the related literature reports are rare.This study identified the expression of BRCA1 and 53BP1 protein among HT with PTC,HT and TND through Polink-2 plus Polymer HRP Detection system and discuss the function of these protein in thyroid tissue,so as to provide experimental basis for clinical.Methods:Drew material from wax sample from January 2013 to August 2015 in hebei province People’s hospital of General subject that confirmed by surgery pathology,24 HT with PTC,24 HT,25 TND then detect the expression of BRCA1 and 53BP1 through immunohistochemical method(PV).Computer image analysis semi-quantitative System was used to analysis the immune positive staining of the subject by average optical density(AOD).All date were presented as mean±standard deviation,the statistical analysis were described by SPSS13.0 software with One-way ANOVA with-group t-test between-goup.P<0.05 for difference of stastistical significance,P<0.01 as the ia a significant difference of statistically significant.Result:Immunohistochemical results showed that weak BRCA1 expressed in the most areas of HT with PTC group,HT group and TND group except some scattered weak expression within some proliferating area of thyroid follicles.The expression rate of 53BP1 among HT with PTC group was 70.8%(17/24),HT group was 58.3%(14/24),and TND group was 16%(4/25).Conclusion: 1.BRCA1 rarely expressed in HT with PTC group,HT group and TND group except some scattered weak expression within some proliferating area of thyroid follicles,suggesting that BRCA1 may play a role in the proliferation of thyroid tissue.2.53BP1 is highly expressed in HT with PTC,HT goups,less expressed in in TND group.suggesting an stronger actvation of DDR in HT with PTC,HT groups.PV can be used to detect the expression of 53BP1 in the HT with PTC,HT,and TND thyroid tissue and be used to determine the thyroid gland of the gene GIN. |
