Objective:To explore the animal model manufacture method of Congenital muscular torticollis.,and use the tuina to treat this kind of animal model.Expect to find one manufacture method of animal model which need the short manufacture cycle and also have the similar pathology change with the CMT, and research the Action mechanism of tuina based on this animal model.lay the foundation for the clinical development CMT multi-disciplinary research.Methods:Healthy male and female rabbits 48,weighing450-550g,were randomly divided into ligation of external jugular veincontrol group (A group) 12;injection of absolute alcohol group(B group)12;injection of hemorrhoid injection (C group)12;control group(D group) 12.Prolifer-ation degree of fibrous tissue were observed by Hemato-xylineosin staining.Expression of alpha smooth muscle actin (a-SMA) in these specimens was detected by immunohistochemistry and computer image evaluation. Then divided healthy male and female rabbits 24 into 3 models:model group 1,model group2 and tuina group.Each group use the idel animal model manufacture method which was be developed in the earlier research. The specimen of model groupl and model group 2 will be take after 1month,2 months separately.Each group was detected by HE test and a-SMA immunohistochemistry.Result:1. The 15th days examination result:HE examination shows that the B group's textile fiber proliferation degree is bigger than A and C two groups obviously.Only B group was positive for a-SMA.2. The 30th days examination result:HE examination shows that the three model groups appear the textile fiber proliferation, but their textile fiber proliferation degree has the big difference(P<0.05), may obtain according to three groups of average order, proliferation degree of the B group is bigger than A and C two groups obviously. a-SMA immunohistochemistry examination shows that the three model groups appear the positive expression for a-SMA,but their positive rate are unequal (P<0.05). Shows after the LSD multiple comparisons, there have the difference between three group of each other, may think B group>A group>C group according to the average value of the positive rate of each group;D group has no positive expression.3. The result of HE examination:the textile fiber proliferation degree of the tuina group is smaller than two model groups obviously (P<0.05).4. The result of a-SMA examination:B group have no positive expression for a-SMA.Conclusion:1.Three methods may manufacture the CMT animal model.2.The absolute alcohol injection group's has fiber structure proliferation compares other two methods to be more obvious, the time which appears also early.3. Tuina may reduce the textile fiber proliferation degree of muscular torticollis rabbit model effectively. |
