Effects Of The Traditional Chinese Medicine With Functions Of Invigorating Qi And Tonifying Kidney On Monoamine Neurotransmitters In The Brain Tissue Of Diabetes Mellitus Rats And Diabetes Cerebral Ischemia Rats

Objective:To study the effects of the Traditional Chinese Medicine with functions of Invigorating Qi and Tonifying Kidney on monoamine neurotransmitters such as norepinepbrine (NE),dopamine (DA) and 5-hydroxytryptamine (5-HT) in the brain tissue of diabetes mellitus rats and diabetes cerebral ischemia rats. Methods:Experiment 1:The diabetic model of Wistar rats was constructed with intraperitoneal injection of streptozotocin.Then the model rats were randomly divided into 2 groups:model group,and TCM treated group.Taking the healthy rats were faton as normal control group. The rats of the TCM treated group were lavaged with 600mg/kg (kg-d) naoshenkang(which is the Traditional Chinese Medicine with Function of Invigorating Qi and Tonifying Kidney), and the rats of the model group and the normal control group were given equal amount of normal saline. All rats were killed on the sixth weekend, and the level of blood sugar, insulin and the monoamine neurotransmitters in the discrete brain areas of rats were determined. Experiment 2:Grouping and modeling as the experiment 1. After feeding six weeks, all rats were given intraperitoneal, anesthesiaed with 3.5 ml/kg of 10% Chloral Hydrate. the model of cerebral ischemia were fuilt by bilateral carotid artery ligation of the rats of Diobetes. All rats model group and TCM treatment group Blank group were treated the same as the surgery group, but without bilateral carotid artery ligation. the preparation, collection of serum and brain tissue and the determination of monaarmine neutransmitters were same ad experiment 1. Results:Experiment 1:The level of blood sugar and insulin of TCM treated groups were significantly lower than that of the model groups(P<0.05), the level of norepinephrine (NE), dopamine (DA) in TCM treated groups descended while the 5-hydroxytryptamine (5-HT) in that increased significantly. There were of the existed between of the Monoamine Neurotransmitters significant different between TCM treated and model groups (P<0.05). Experiment 2:The level of blood sugar of TCM treated groups were significantly lower than that of the model group and blank group (P<0.05), and ISI of TCM treated groups rised obviously than thas of the blank and model groups,but IR of arlicin group descended obviously than that of blank and model groups (P<0.05). The level of NE, DA,5-HT in model group were significantly lower than that of the blank group (P<0.05), but in TCM treated groups was higher than that of the model group (P<0.05). There is no statistics difference between TCM treated and blank group (P>0.05). Conclusion:The Traditional Chinese Medicine with functions of Invigorating Qi and Tonifying Kidney have the effects of raising ISI,improving IR and lowering blood sugar, and also can efficiently regulate monoamine neurotransmitters in brain tissue of diabetes mellitus cerebral ischemia rats.
