ObjectiveTo investigate the expression of tumor suppressor gene maspin and casepase-3 and MVD in EOC and their relationship with the clinicohistological features of EOC.To explore the potential role of maspin in the development of EOC.MethodsExpression of maspin and casepase-3 and MVD in 50 cases of EOC,20 cases of benign tumor and 15 cases of normal ovary tissue. Diagnosed by the affiliated hospital of Taishan Medical University and the Military Hospital of Jinan during Jan 2000 and Dec 2009 were detected immunohistochemically. The average age of the 50 cases of EOC is 56. The 50 cases of EOC includes 29 serous cystadenomas of ovary,12 mucous cystadenomas of ovary and 9 endometrioid carcinomas; 12 stageⅠ,10 stageⅡ,16 stage III and 12 stageⅣby FIGO staging;8 G1,12 G2 and 30 G3 by differentiation.Statistical analysis was performed with statistical program for social sciences 13.0 software,using Chi-square,one-way ANOVA, t-text and nonparametric test.Statistically significant level was considered as "alpha equals 0.05".Results1. The positive rate of maspin in 50 cases of EOC was 52.0%, in 20 cases of benign tumor was 25.0%, in 15 cases of normal ovary tissues was 20.0%. The expression of maspin in EOC and in the benign tumors was significant differences as well as in EOC and in normal ovary tissues (P<0.05, P<0.05). The expression of maspin was no significant differences in the benign tumors and in normal ovary tissues (P>0.05). The expression of maspin in EOC is related with the histological grade, FIGO staging and lymphnode metastasis, (P<0.05), but the expression is no related with the age,non-ascites and pathological type (P>0.05).2.The positive rate of caspase-3 in 50 cases of EOC was 56.0%,in 20 cases of benign tumor was 25.0%,and in 15 cases of normal ovary tissues was 26.7%. The expression of caspase-3 was significant differences between EOC and the benign tumors (P<0.05).The expression of caspase-3 was no significant differences in the benign tumors and in normal ovary tissues (P>0.05). The expression of caspase-3 in EOC is related with the histological grade and FIGO staging, (P<0.05), but the expression is no related with the age, pathological type, non-ascites and lymph node metastasis (P>0.05).3. The number of MVD in 50 cases of EOC was 16.67±3.2, in 20 cases of benign tumor was 8.40±2.33, in 15 cases of normal ovary tissues was 8.47±2.59. The expression of MVD in EOC and in the benign tumors were significant differences as well as in EOC and in normal ovary tissues(P<0.05,P<0.05). The expression of MVD was no significant differences in the benign tumors and in normal ovary tissues (P>0.05).The expression of MVD in EOC is related with the histological grade, FIGO staging and lymphnode metastasis,(P<0.05), but the expression is no related with the age,non-ascites and histologictype (P>0.05).4. The expressions of maspin and caspase-3 had positive correlation in the same samples(rs=0.439, P<0.01).5. The expressions of maspin and MVD had negative correlation in the same samples(rs=-0.405, P<0.01).Conclusion1. The expression of maspin is up-regulation in the normal ovary tissues, benign tumor and the EOC, but the expression of maspin in EOC is related with the histological grade, FIGO staging and lymphnode metastasis (P<0.05), which indicts that the expression of maspin is related with carcinogenesis, develop-ment, invasion, metabasis and prognosis of EOC.2. The expression of caspase-3 is up-regulation in the normal ovary tissues, benign tumor and EOC,and the expression of caspase-3 is significant differences between EOC and the benign tumors (P<0.05), which indicts that the expression of caspase-3 is related with carcinogenesis and development of EOC. 3. The expression of MVD is up-regulation in the normal ovary tissues, benign tumor and EOC, and the expression of MVD is significant differences between EOC and the benign tumors (P<0.05), which indicts the expression of MVD is related with carcinogenesis and development of the EOC.4. The expressions of maspin and caspase-3 have positive correlation in EOC, which means both of them have synergistic effect.The maspin may promote the apoptosis of the EOC by enhancing the founction of caspase-3. So that it can restrain the carcinogenesis and development of the EOC.5. The expressions of maspin and MVD have negative correlation in EOC, which means the maspin may restrain the carcinogenesis, development, invasion and metastasis of the EOC by restrain the angiogenesis of the EOC.
