Betulin has toxicity less and are rich in resources etc. various advantages. It's pharmacological effects mainly displays in anti-inflammatory, antivirus and antitumor function, etc.It still can cleare free radicals by improving of the activity of enzyme, scavenging free radicals In the body. Restrain free radical mediated lipid peroxidation of liver damage. Consequently to protect liver cell structure and function. Improve the functions of body dependence antioxidant enzymes system. But the effect of betulin in radiation damage research has yet to see reports.Objective:Study the therapeutic effect of mice and its mechanism from pharmacology aspects of betulin X-ray damage through the establishment of X-ray damage model. Methods Mice were randomized six group, they are solvent control group, model group, positive drug group, low dose group, middle dose group, high dose group etc. Every groups are gave One-time X-ray systemic exposure to Establish the radiation damage model except solvent control group. After 10 days later calculation change of weight with mice during the experiment. Counting Peripheral leucocyte(WBC) use white blood count method on the fourth and tenth day, and counting the marrow is nuclear cell count (BMNC) on the tenth day. Calculating the index of thymus and spleens, Counting the number of Stem colony-forming uints (CFU-S)in spleens. Detecting SOD activity in serum, bone marrow, liver, kidney use the method of adjacent pyrogallol, and detecting the contents of MDA use the method of TBA. Detecting the content of BUN, Cr, ALT, AST in serum.Result:1. The influence of general situation of betulin to miceAfter X ray, each illuminate group of mice weight were significantly lower than the control group. After the treatment, MD and HD betulin can make X-ray damage mice weight increased significantly (P<0.05).PG and LD compared with model group.mice weight change has no significance difference. 2. The influence of WBC of betulin to miceUse white blood count method to counting WBC which were from after X-ray, 4day later and 10days later.Results:After X-ray, compared with CG,each illuminate group WBC count were significant lower than CG (P<0.05). Among each illuminate group, WBC count has no significant different. Imply that X-ray damage model manufacturing success. After X-ray 4days betulin and Huangqi injection have no effects in increasing WBC count in mice. After X-ray 10days, betulin of MD and HG could increasing the number of WBC count in mice (P<0.05), Huangqi injection and betulin of LD were have no significant difference compared with MG (P>0.05). Imply that betulin with MD and HD could improve the number of WBC count.3. The influence about the number of NBMC of betulin to miceUse white blood count method to counting the number of NBMC in mice. Results: The number of NBMC in mice of MG and PG were lower than CG (P<0.05), The number of NBMC in mice of LD, MD,HD were have no significant difference compared with CG (P>0.05). The number of NBMC in MD and HD were higher than MG (P<0.05). Imply that betulin could improve the bone marrow inhibition which cause by X-ray X-ray.4. The influence about the thymus and spleen index of betulin to miceAfter the end time give medicine 30min to executed mice, remove the thymus and spleen to weighing. Calculation thymus index and spleen index of mice.Results:Thymus index has no significant differences among each group (P>0.05). All illuminate groups spleen index were lower than CG (P<0.05).and there were no significant differences among each illuminate groups (P>0.05). Imply that betulin and huangqi injection were have no significant effect about improve thymus index and spleen index.5. The influence of betulin about content of MDA in miceResult:MD and CG were have less MDA in serum than MG (P<0.05). there were no significant differences among other groups about MDA in serum. LD and CG were have less MDA in Bone marrow than MG (P<0.05). here were no significant differences among other groups about MDA in Bone marrow. Content of MDA in liver and kidney were no significant differences among each groups (P>0.05)Imply that betulin maybe have the effect of reduce the content of MDA in serum and bone marrow, but it didn't show obvious function in reduce the content of MDA in Liver and kidney.6. The influence of betulin about active of SOD in miceResult:Huangqi injection and betulin were didn't show obvious function in the aspect of improving the activity of SOD in serum. The activity of SOD in bone marrow were higher in PG, LD and MD than MG (P<0.05), The activity of SOD in HG was higher than in MD (P<0.05). The activity of SOD in liver in HD and CG were higher than MG,PG and LD (P<0.05), The activity of SOD in kidney in LD,MD and HG were higher than MG (P<0.05),and MG was higher than PG,LD and HD (P<0.05).but it had no significance difference compared with CG (P>0.05). The result imply that betulin have no effect in improve the activity of SOD in serum, but it show different degree of functions in improving the activity of SOD in bone marrow, liver and kidney. Huangqi injection could improve the activity of SOD in bone marrow.7. The influence of betulin about serological index in miceUse automatic biochemistry analyzer detection the content of BUN, Cr, ALT, AST in serum. Result:Hiangqi injection and betulin could reduce the content of BUN. ALT, AST in serum. Betulin of LD and MD could reduce the content of AST, it has no effect in reducing Cr.8. The influence of betulin about CFU-S in miceUse spleen nodules method to count CFU-S in spleen. Result:CFU-S in PG, MD and MD were higher than MG (P<0.05). There is no significant about CFU-S among other groups. The result imply that betulin and huangqi injection were could improve CFU-S in mice, improve the damage of hematopoiesis and immune loss which caused by x-ray.Conclusion:1.Betulin could improve WBC in mice which damaged by X-ray. Improve the inhibition of BMNC, increase CFU-S in mice. It has no significant effect in improving thymus and spleen index. 2. Betulin could enhance the activity of SOD in bone marrow, liver and kidney, and it could reduce the content of MDA in serum and bone marrow. But it has no definite effect in reducing the content of MDA.3. Betulin could reduce the content of BUN,ALT,AST in serum, but it has no significant effect on reducing the content of Cr.4. It imply that betulin have certain curative effect on X-ray damage mice, it's mechanism be that it could reduce free radicals in organism. |
