| Objective:Our aims are to study the effect of Jingqianshu granule on expression level of ERβand related proteins, study the effect of Jingqianshu granule on corpuscular viability of neonate rat hippocampal neurons cultured in vitro, seek for parts of the mechanisms of Premenstrual Syndrome liver-qi depression; and study the effect on Jingqianshu granule acting on the liver-qi depression.Methods:Before the study, we observed the vagina cell shape to make sure the estrum of rats:receptive phases and non-receptive phases, selected the non-receptive phases rats, and before we took the serum we estimated model rats by open-field test.Premenstrual syndrome (PMS) model rats with liver-qi depression were prepared by binding method, took Jingqianshu granule medicated serum, cultured neonate rat hippocampal neurons and assay the viability of neural cells by MTT; the expression levels of ERβ,5-HTR1 A. MAOA,MAOB,5-HTT in the neurons were evaluated by western blot and RT-PCR.Results:Compared with the blank group rats, the body weight of the Premenstrual syndrome (PMS) model rats decreased significantly, compared with the Premenstrual syndrome (PMS) model rats, the body weight of the medication administration group rats increased significantly (P<0.05). Compared with the blank group, the crossing score, the rearing score and the total score of the PMS model rats in open-field decreased significantly (P<0.05), compared with the PMS model rats, the scores of the medication administration group rats in open-field increased significantly (P<0.05). According to the results of MTT, Jingqianshu granule medicated serum could markedly increase the viability of neonate rat hippocampal neurons. According to the results of western blot, it showed that compared with the nomal hippocampal neurons, the ERβexpression levels of hippocampal neurons cultured with Premenstrual syndrome (PMS) model rats serum increased significantly (P<0.05), compared with hippocampal neurons cultured with PMS model rats serum, the ERβexpression levels of hippocampal neurons cultured with Jingqianshu granule medicated serum decreased significantly, the MAOA expression levels showed the same results with ERβ; while compared with the nomal hippocampal neurons, the 5-HTR1A expression levels of hippocampal neurons cultured with Premenstrual syndrome (PMS) model rats serum decreased significantly (P<0.05), compared with hippocampal neurons cultured with PMS model rats serum, the 5-HTR1A expression levels of hippocampal neurons cultured with Jingqianshu granule medicated serum increased significantly, the expression level of 5-HTT showed the same results with 5-HTR1A; the expression of MAOB showed no statistical significance. |
