Breast cancer is one of the most common malignant tumor in women,whose incidence is 7~10% in all kinds of malignant tumor.Breast cancer has been the main threat to the women's health after metrocarcinoma.The pathogenesis of breast cancer is not clear at present.Estrogen before and after menopause is the obvious factors to happened on breast cancer.Besides,heredity,diet,rationalized factors and some benign breast disease have something to do with the happened on breast cancer.On our opinion,the pathogenesis of breast cancer is HMEpiC have genetic mutations under many factors of cancer,resulting in cells growth out-of-control.Since the biologic behavior of cancer cells have been changed,there is disorder and unlimited malignant hyperplasia.In histology,it shows massive immature cancer cells proliferate infinitly and huddle out-of-order,which press and erode to destroy normal structure around in breast.It has been confirmed the interaction between many chemotactic factors and their receptors play an important role in the evolve of the breast.Chemotactic factors and their receptors have been hot spots in the emergence and development of breast.In 2001,Muller reported research findings about SDE-1—CXCR4 system, CLL2—CCR7 system and the breast at first.After that,there is many researches about the correlation between IL8—CXCR1/CXCR2 system, RANTES—CCR5 system and breast. Part I Expression of the chemokine receptor 5 in breast invasiveductal cancinoma and metastasized axillary lymph node ObjectiveTo study the expression of chemokine receptor 5(CCR5) in breast cancer and metastatic axillary nodes, and explore the effect on the pathogenesis of human breast cancer.Methods35 samples were collected from department of Breast & Thyroid surgery, Liaocheng Peoples'hospital from Aug,2008 to Apr,2009,pathology results of which were breast invasive ductal carcinoma, among which 28 patients with axillary lymph node meta-stasis.At the same time,33 samples of axillary lymph node were collected, in which were 20 cases confirmed pathologically breast cancer metastasis.According to UICC,there were 12 cases(stageⅡ).21 cases(stageⅢ),2 cases(stageⅣ).To also collected 35 cases of normal tissues and 21 benign breast tumor tissue as control.Reverse transcriptase polymerase chain reaction (RT-PCR) were used to detect the relative expression quantity of CCR5mRNA in pri-mary breast cancer foci,its axillary lymph node,PCHT and fibroadenoma of breast. The Polymorphism of CCR5-δ32 was tested by DNA electrophoresis.Results1.The expression of CCR5mRNA in the groups of breast tissue:the expression of CCR-5 mRNA in breast cancer is significantly higher than in adjacent normal tissues and in breast fibroadenoma tissues(P<0.05). The relative expression quantity of CCR5 mRNA is different between pathologic stagingⅡandⅢin statistics (P<0.05), but no different between adjacent normal tissues and breast fibroadenoma tissues (P>0.05)2. The expression of CCR5mRNA in the groups of axillary lymph node:the relative expression quantity of CCR5 mRNA in metastatic axillary nodes is significantly higher than in no-metastatic nodes (P<0.05)3. Primary tumor tissue of breast cancer CCR5mRNA expression and immunohisto chemical indicators of clinical relevance:there is correlated relationship between CCR5 mRNA with axillary lymph node metastasis and breast oncogene C-erB-2, but has no correction with other immunological and biochemical indicator.Conclutions1.CCR5mRNA was expressed significantly higher in breast cancer tissue than paracar-cinoma tissue and breast fibroadenoma tissue. Ligand-receptor interaction between CCR5 and its ligands may be played a role in the progress of breast cancer.2.We found that CCR5 mRNA in breast cancer tissue was expressed significantly high-er in GreadⅡ(UICC) than GreadⅢ(UICC).Ligand-receptor interaction between CCR5 and its ligands was promoted the progress of breast cancer.3.We detected higher expression of CCR5 mRNA in metastatic axillary nodes than no-metastatic nodes,so CCR5 may be correected with the progress of axillary lymph node metastasis in breast cancer.4.The interaction between CCR5 and its ligands plays an important role in carcinogene-sis, envolvment and axillary lymph node metastasis of breast cancer. CCR5 will be significantly molecular indicator to predict axillary lymph node metastasis and in targeted therapies.ObjectiveTo study the frequency of polymorphism CCR5-832 in breast cancer and metastatic axillary nodes, and explore the effect on breast cancer metastasis.Methods105 samples were collected from department of Breast & Thyroid surgery, Liaocheng Peoples'hospital from Aug,2008 to Jun,2009,pathology results of which were breast invasive ductal carcinoma, among which 87 patients with axillary lymph node meta-stasis.At the same time,50 samples of axillary lymph node were collected, in which were 34 cases confirmed pathologically breast cancer metastasis.According to UICC,there were 5casea(stageⅠ),4lcases (stageⅡ),53 cases(stageⅢ),4cases (stageⅣ).To also collected 105 cases of normal tissues and 40 benign breast tumor tissue as control. The Polymorphism of CCR5-δ32 was tested by DNA electrophoresis.Results1.The incidence of CCR5-Δ32 in breast tissue:there is not CCR5-Δ32 allele in breast cancer, adjacent normal tissues and breast fibroadenoma tissues 2. The incidence of CCR5-Δ32 in are palpable mobile axillary nodes organization:Whether the are palpable mobile axillary nodes organization in breast transfer or not,there is not CCR5-Δ32 allele.ConclutionsThere is not CCR5-Δ32 allele in breast organization and are palpable mobile axillary nodes organization.At present, this research does not confirm CCR5-Δ32 allele promote the process of invasive ductal carcinoma of the breast from happened to development.It does not confirm CCR5-Δ32 allele has something to do with axillary nodes metastasis in IDC.The role CCR5-Δ32 allele plays in breast cancer is worth further studying.
