OBJECTIVE:1,To detect the changes of ADMA,MDA and Ca2+ in hypoxia cells treated by Shuxuening in vitro,so to explore the machenism of improvment of endothelial cells function by Shuxuening.2,To evaluate the changes of serum Asymmetric dimethylarginine, malondialdehyde,homocysteine and C reactive protein levels of cerebral infarction patients before and after treated by shuxuening, so to explorethe mechanism of Shuxuening in treatment in cerebral infarction.METHODS1. Fresh umbilical cord vein endothelial cells were cultured to establish hypoxia models. Cells were divided into two groups by different drugs: Shuxuening group and the control group, and cells without hypoxia treatment were to be normal group. Cells relative number and relative activity were evaluated by MTT assay; intracellular Ca2+ concentration was detected by Ca2+ fluorescence staining; ADMA levels was detected by high-performance liquid chromatography; changes of serum MDA was detected by the thiobarbituric acid method; Apoptotic cells were detected by PI staining. Mechanism of Shuxuening was analyed by the above research results.All data were analyzed by SPSS 13.0 software.2.96 cerebral infarction patients selected in our hospitals were divided into two group:37 patients with general treatment and 59 patients with Shuxuening treatment.32 more patients were selected to be as normal control. Serum ADMA levels, hs-CRP and Hcy levels in the divided there groups were detected by high-performance liquid chromatography and immunoturbidity method:serum MDA was detected by the thiobarbituric acid method; reduced infarct size before and after treatment were measured; these therapeutic effects were analyzed. All data were analyzed by SPSS13.0 software.RESULTS1. OD value of Shuxuening group was significantly higher than the saline control group (P<0.05); ADMA,MDA levels in the control group cells cultured in the Economic and hypoxia was significantly higher than the normal group after hypoxia ADMA,MDA levels (P<0.05);The ADMA,MDA levels in Shuxuening group cells treated by hypoxic culture were significantly lower than that of the control group(P<0.05);The Ca2+ fluorescence intensity levels in Shuxuening group cells treated by hypoxic culture was significantly lower than that of control group (P<0.05); The red fluorescence density by PI staining in Shuxuening group cells treated by hypoxic culture was significantly lower than that of control group (P<0.05).2,Serum ADMA,Hcy and hs-CRP levels were markedly higher in acute phase of cerebral infarction group than normal control group before treatment (P <0.05). After treatment, there were significant difference of the serum Hcy and hs-CRP levelsbetween shuxuening groupand the control group, (P<0.05). After shuxuening treatment, the reduced infarction size was significantly larger than that of general treatment group (P<0.05).CONCLUSION1. ADMA release and synthesis in the endothelial cells by hypoxia culture in vitro was reduced by Shuxuening treatment; Shuxuening could inhibit calcium overload, reduce damage to cells by free radical, reduce cell death, protect the vascular endothelial cell function and reduce the occurrence of atherosclerosis. It may be one of the mechanism of it can prevent and cure the ischemic cerebrovascular diseases.2. There was a significant effect of Shuxuening on cerebral infarctionand a greatly reduction in infarct size. These is maybe related to the function of Shuxuening to clear free radicals and reduce serum ADMA, HCY and hs-CRP levels.
