| Objective:To research the effect of Polysaccharides GF extracted by Ganoderma applanatum(Per.ex Gray ) on cell telomerase activity of tumor-bearing mice and cyclin dependent kinases2 gene expression.To Investigate the mechanism of GAPSGF anticancer by molecular biological level.Method:There are four groups of in the experiment: One group is the normal control group, the other three groups are negative control group,positive control group and GAPSGF group after establishment of mouse H22 portability hepatoma model.GAPSGF group was given the medicine according to 50mg/kg·d-1,positive control group was given cyclophosphamide according to 30mg/kg·d-1,normal group and negative control groups were given normal saline,until the negative control group mice tumor achieves above 1g, then execute the mice the next day.Then peel entire tumour tissue of every group, weight,calculate tumor inhibit rate. Detect cell telomerase activity of tumor-bearing mice and cyclin dependent kinases2 gene expression by ELISA.Result:1.Compared to tumor weight of negative control group,tumor weight of GAPSGF group and positive control group are lower, there are significant differences between them(P<0.05).2.Cell telomerase activity expression in GAPSGF group and positive control group are lower than it in negative control group( P<0.05).And there are no significant differences between GAPSGF group and positive control group (P>0.05).3.Compared to negative control group, cyclin dependent kinases2 gene protein expression obviously are lower in other three groups( P<0.05).There are significant differences among any two groups(P>0.05).Conclusion:1.GAPSGF had obvious tumor lower effecton H22 hepatoma tumor mice.And tumor inhibit rate was 42.25%.2.GAPSGF can reduce cell telomerase activity of tumor bearing mice obviously.3.GAPSGF can reduce cyclin dependent kinases2 gene expressi- on,and the dose of it approachs the dose of normal group. |
