Objective:To investigate the protective function of ganoderma applanatum polycose against gastric ulcer and its mechnism.Methods:①We prefer 50 Wistar rats for our object with male and female each half. Constraint-stress gastric ulcer model and pylorus ligation gastric ulcer model were applied to discuss the protection of ganoderma applanatum on gastric ulcer.The constraint-stress rat gastric ulcer model was duplicated and 50 rats were randomly divided into five groups with each 10:nomal group,model group,ganoderma applanatum polycose 0.25g/kg group,ganoderma applanatum polycose 0.5g/kg group and ganoderma applanatum polycose 1.0g/kg group.The constraint-stress rat gastric ulcer model was performed 2 hours after the last administration of ganoderma applanatum polycose,the rats underwent anesthesia with ether,fixed their extremities on the iron net,and with their head-down ward vertically for 16 hours(except the normal group).Then the rats were administered with pentobarbital and cut the rats' belly open to get the stomach and obtain blood through abdominal aorta in order to determine the concerntration of malondialdehyde(MDA),reduced glutathione hormone(GSH),nitrogen monixidum(NO) and superoxide dismutase(SOD) activity of gastric mucosa and blood serum as well as evaluate the injury index of rats' gastric mucosa.②The pylorus ligation rat gastric ulcer model was duplicated and 80 rats with male and female each half were randomly divided into four groups with each 20: model group,ganoderma applanatum polycose 0.25g/kg group,ganoderma applanatum polycose 0.5g/kg group and ganoderma applanatum polycose 1.0g/kg group.2 hours after the last administration of ganoderma applanatum polycose,10 rats of each group were cut the belly open under anesthesia to ligate pylorus,150mmol/L saline 4ml were injected into the stomach via duodenum and pylorus.4 hours after pylorus ligature, they were decapitated to death and cut the belly open to get the stomach,clippered the stomach along the lesser gastric curvature and got the gastric juice collected. Determine the volume of dissociated mucosa in the stomach and to pick the free mucosa accreted to the surface of gastric mucosa.Soak the stomach in the solution for 2 hours,then took the stomach out,reactive liquid certrifugated and determine the volume of gastric mucosa.18 hours after pylorus-ligature,the rest rats were decapitated to death,cut the belly open,ligatedcardiac orifice,get the stomach and collecte gastric juice.The volume of gastric juice was recorded as well as determine the activity of total gastric juice,free acid and pepsin and evalute the injury index of the rats' gastric mucosa.Results:①Compared with the model group,Ganoderma applanatum polycose of middle and high dosage could decrease the gastric grade(P<0.01) by 44.65%and 54.05%respectively;Ganoderma applanatum polycose of middle and high dosage could increase the SOD activity of rat's serum and gastric mucosa as well as rat's serum GSH concerntration rising from stress(P<0.05,P<0.01),while decrease the MDA concerntration of gastric mucosa(P<0.05);Ganoderma applanatum polycose of any dosage could decrease MDA concerntration of rat's serum(P<0.05,P<0.01).②Compared with model group,Ganoderma applanatum polycose of any dosage could decrease the gastric grade of pylorus ligation rats(P<0.05,P<0.01) by 19.30%,24.95% and 56.34%respectively;It also increased the concerntration of gastric mucosa secretion and decreased free acid and total gastric acidity,as well as decreased pepsin activity(P<0.05),but with no differences of the gastric volume.Conclusion:Ganoderma applanatum polycose presents to be of anti-experimental gastric ulcer and the mechanism is probably related with the increasingly release of GSH and NO in plasm and gastric mucosa,enhencement of SOD activity,decrease of MDA concerntration and improvement of gastric mucus barrier function as well as inhibition the activity of gastic acid and pepsin.
