Objectives1,Transplante the MMP-9 gene silencing BGC823 cells and BGC823 cells into nude mice, and built orthotopic tumor model.2,Study the impact caused by the MMP-9 gene silencing or non-silent BGC823 cells in nude mice tumor formation rate, tumor growth and size, volume and transfer, etc.MethodsRandomly divided the 42 nude mice into 5 groups: The first group was inoculated MMP-9 gene silencing BGC823 cells in the stomach, referred as the stomach-positive group, a total of 10; second group inoculated MMP-9 gene is not silent BGC823 cells in the stomach, referred as the stomach-negative group, a total of 10; third group was inoculated with normal saline in the stomach, referred as the saline group, a total of 10; fourth group was inoculated MMP-9 gene silencing BGC823 cells in the right armpit, referred as the armpit-positive group, a total of six ; fifth group inoculated MMP-9 gene is not silent BGC823 cells in the right armpit, referred as the armpit negative group, a total of six. By ether anesthesia, opened the abdominal cavity and locate the stomach, inoculated mucosa MMP-9 gene silencing BGC823 cells and BGC823 cells and normal saline in the greater curvature side of gastric. And then inoculated MMP-9 gene silencing BGC823 cells and BGC823 cells in the right axillary subcutaneous. After one month, killed nude mice in the SPF animal laboratory under the conditions of rearing to observe tumor formation rate, tumor growth and size, volume and transfer case, parallel HE dyeing.Results1,Rate of the growth of tumors and the size and volume of tumor: armpit vaccinated group and the composition of in situ tumor inoculation rate of 100%, no statistical significance, no saline group into the tumor, axillary-negative tumor size and volume of the composition of positive group are larger than which of the armpit and statistically significan(tP<0.05);stomach-negative tumors the size of the composition, size and stomach-positive group have no difference, no statistical significance. 2,Tumor metastasis: The armpit vaccinated group and saline group were no intra-abdominal, chest and transfer. The stomach-positive group and negative group laparotomy and the stomach exploratory thoracotomy, the stomach-positive group did not find the transfer of abdominal cavity and chest, but the stomach spleen negative group found that two spleen transfers, two intra-abdominal metastasis, one liver metastasis, but lung metastasis was not found. Stomach-positive group and negative group stomach metastasis statistical significance(P<0.05).Conclusions1,Nude mice model of orthotopic transplantation in gastric wall was successfully constructed for the future construction of animal model of orthotopic transplantation and application one of opening up important avenues.2,RNAi-mediated gene silencing of MMP-9 can significantly affect the BGC823 cells in nude mice tumor growth and distant metastasis. MMP-9 is expected to become a new target for the treatment of gastric cancer.
