| Objective:To investigate the inhibitory action of small molecular peptide from Centipede and its mechanism on nude mice model of human hepatocellular carcinoma by using orthotopic transplantation.Method:1. To use the Enzyme-Decomposition method obtained the crude extracting of Centipede which was injected into nude mouse after separated and purified by using the Sephadex G-25gel chromatography2. Chose five nude mice were given to normal groups among the sixty nude mice with random selection, to establish the numbers of fifty-five nude mice models with human hepatocellular carcinoma by orthotopic transplantation and the forty-eight of those models were succeed and were randomly divided into4groups which are Model Group, high dosage of the SMPC, low dosage of the SMPC and control group Of5-FU. Each treated groups were applied by intraperitoneal injection, according to the high dosage of the50mg/kg/d SMPC, low dosage of the10mg/kg/d SMPC, control group Of the50mg/kg/w5-FU and those in Model group were given normal saline instead of SMPC.3. After fourteen days dosed, peeled off the tumor tissues and weighted them, calculated the tumor-inhibition rate, took the blood from the orbital veins of nude mice for the assay and determined the serum concentration of AFP. In the meantime, the protein expressions of Ki67〠EphA7and EphB1in the tumor tissues were examined by LASAB immunohistochemical staining method.Result:SMPC had the inhibitory effects on the growth of nude mice models with human hepatocellular carcinoma by orthotopic transplantation.The average of tumor weight with the high dosage of the SMPC and low dosage of the SMPC were Significantly and respectively lower than the average of tumor weight with Model Group (P<0.01).And the average of tumor weight with high dosage of the SMPC was the lowest one compared the others (P<0.01). Simultaneously,the same results were both happened in determining the serum concentration of AFP and the examining the expression of Ki67.The only difference is these results of control group Of5-FU was higher than the one of high dosage of the SMPC (p<0.05). Moreover, both the high dosage of the SMPC and the low dosage of the SMPC were significantly activated the express activity of EphB1in the tumor tissue but inactivated the express activity of EphA7compared the control group of5-FU.Conclusion:1. In this paper, the obvious inhibitory action of small peptide from Centipede was tested on nude mouse model of human hepatocellular carcinoma by using orthotopic transplantation.2.SMPC has inhibited the growth and proliferation of tumor cell as its’mechanism of action through intervening the signal transduction of tumor cell, regulating tyrosine by phosphorylation, activating the express activity of EphB1and inactivating the express activity of EphA7. |
