Objective To study the dynamice expression of AQP-4 in cerebral ischemia-reperfussion model in rats and the influence of the injection of Radix Salviae Miltiorrzhizae and Ligustrazin.Methods 100 healthy Sprague-Dawley rats were randomly allocated into low-dose drug-therapy(n=25),high-dose-drug-therapy(n=25),control(n=25),sham-operation(n=2 5)group.The rats in all the groups were redivided into 5 subgroups according to the different time periods of brain ischemia, All the rats were tested for Longa neurological behavior,then killed by decapitation and the brains were taken out at 6 hours,24 hours,72hours,5days and 7days after establishment of the model. Brain water content was measured by the wet-dry method and we observed the pathology changes by hematoxylin and eosia(HE) dyeing,the expression of AQP-4 by immunohistochemistry staining.Results①the behavioral outcomes were most serious at 72 h;②The brain water content(BWC)began to increase in the ischemic side of hemisphere at 6 hours after cerebral infarction, it reached the peak at hour 72, and then decreased gradually, but it was still higher than that in the sham-operation group at day 7-There was statistical significance during the contral group,low-dose therapy and high-dose therapy at different time points (F=137.23,F=63.16.F=22.42,each P< 0.01)。The BWC in the infarct hemisphere in the therapy and control groups were obviously higher than that in the sham-operation group at every point of time(all P< 0.01),and that of therapy group were lower than the control group(all P<0.01),and that of the high-dose therapy group were lower than the low-dose therapy group(all P<0.01)。③The level of AQP-4 expression increased in the infarcted side at 6 hours after cerebral infarction, it reached the peak at 72 hours, and then decreased gradually, but it was still higher than that in the sham-operation group at day 7. there was statistic significance during 4 groups at the same time point (each P< 0.01). The AQP-4 expression in the therapy group and the control group were higher than that in the sham-operation group at every point of time(all P<0.01),and the AQP-4 expression in the therapy groups were significantly lower than that in the control group(all P<0.01), and that of the high-dose therapy group were lower than the low-dose therapy group(all P<0.01).Conclusion①The phase consistency is obvious between the change of expression of AQP-4 and dynamic state process of brain edema;②The Injection of Radix Salviae Miltiorrzhizae and Ligustrazin can improve neurological deficits, reduce brain edema after cerebral infarction and inhibits AQP-4 expression;③The Injection of Radix Salviae Miltiorrzhizae and Ligustrazin may reduce cerebral edema thought inhibiting of AQP-4 expression。...
