Objective : The research on the drug Salvia-Safflower cerebral ischemia and reperfusion injury in rat serum BDNF,GDNF factor expression,and to investigate this drug on rats with brain protection mechanisms.Methods: SPF male SD rats using modified Zea-Longa Preparation of focal cerebral ischemia-reperfusion injury(MCAO / R)model,sham group only isolated common carotid,external carotid and internal carotid artery,not Plug.The rats were in the blocking 1.5h,reperfusion after administration 3h.Experimental sham operation group,model group,nimodipine group,SM group,group safflower,Salvia safflower high,medium and low dose group.Drug treatment groups were 10.8mg / kg,1.35 g / kg,0.81 g / kg,3.24 g / kg,1.62 g / kg,0.81 g / kg dose orally,sham operation group and model group at equal volume saline administration.Each group were fed once a day for 2 weeks.After 15 days of treatment,the last administration 2h were drawn.Pharmacodynamic evaluation using HE,Nissl staining morphological changes of brain tissue,using immunohistochemical method,the expression of an enzyme-linked immunosorbent assay in rat serum BDNF,GDNF factor.Results:(1)Neurological function score results: before treatment effects,neurological score model group rats were higher than the sham group,the difference was statistically significant(P <0.05).Effects after treatment: the sham group and the other groups except for the gap in the model group were statistically significant(P <0.05);model group and nimodipine and Danhong high,medium and low dose gap was statistically significant(P <0.05).Danhong high dose group of rats with the rest of the gap between each treatment group was statistically significant(P <0.05).(2)HE staining pathology: each treatment group were obviously pathological structure has been improved,which Danhong high,brain tissue dose group improved the most obvious,a slight reduction in the number of neurons infarction compared with the sham group,the intact cell membrane,nuclear clear,visible necrosis mitigate nerve cell disease have varying degrees of change,organizational structure has been close to normal levels.(3)Nissl staining: neurons sham-operated rats Nissl display up,neurons swelling rat,Nissl model group was significantly reduced or even disappear,and nuclear condensation.Number of Nissl rat each other treatment groups were significantly higher than the model group,neuronal cell morphology tended to be normal,damaged cells are improved.(4)Immunohistochemistry: sham-operated rats hippocampus BDNF and GDNF positive cell count minimum;except SM group and safflower group,the rest of the treated rats BDNF positive cells compared with the model group were statistically significant(P <0.05);rat GDNF positive cells in each treatment group were statistically significant(P <0.05)compared with the model group;Salvia,safflower,nimodipine and Danhong low dose group rats into four groups with the number of positive cells in each other group gaps were statistically significant(P <0.05),but the gap between the four groups was not statistically significant;Danhong high dose group rat BDNF and GDNF positive cell counts higher than other treatment groups,the difference was statistically significance(P <0.05).(5)Enzyme-linked immunosorbent assay: sham group BDNF and GDNF was the lowest;Salvia group,safflower BDNF and GDNF group compared with the other contents of the gap between the groups was statistically significant(P <0.05);Danhong high,high dose group rat BDNF and GDNF were higher than all other treatment groups,the difference was statistically significant(P <0.05).Conclusion:(1)Salvia drug-saffron reduces cerebral ischemia-reperfusion injury in rats the neurological function score improved neurological deficit situation,indicating the drug Salvia-Safflower has a synergistic effect,protect brain function of the rats.(2)Salvia drug-saffron can improve the pathological structure Cerebral ischemia-reperfusion injury in rats,the drug proved to Salvia-Safflower effectiveness of treatment of the disease.(3)Drugs on Salvia-Safflower can enhance cerebral ischemia and reperfusion injury in rat serum BDNF and GDNF factors play a role in brain protection mechanism may be related to this.(4)Drugs Salvia-Safflower high dose for cerebral ischemia-reperfusion injury in rats optimal effect,for the clinical treatment of the disease provided for reference. |
